What he said. Also, there's no problem with contams as the broth cools and air is pulled back inside?
-------------------- Sonic's "I'll buy a pressure cooker someday" Instant Pot times and settings:
- Popping Corn Rapid Re-hydration - Half fill all containers with corn. Empty into Instant Pot. Cover with water. Run 30 minutes on PC. Release, remove, dry.
- Wheat/Rye Berry Soak Re-hydration - Half fill all containers with grain. Empty in pot with water. Soak for 6-12 hours, air dry until surface is completely dry.
- Grain Jar Sterilization - Run 2 - 2.5 hrs on PC
- PastyPlates, MEA Formulation- Run 45 minutes on PC
- LME Liquid Culture - Run 45 minutes on PC

No I screw them tight after inoculation.

I'm going to try this. That looks pretty sweet, Tweeq.

Why do you only drop in a wedge? Not a huge chunk of an entire plate that's been cut up tic tac toe style?
Easier to break up/agitate or just less chance of contam being in a smaller wedge?
I highly recommend the Josex Poke, these ones were tiny wedges of agar though.
Like others say, less chance of contamination. Don't need a big chunk, why bother?

Thanks Mushboy and Asura! I'm not going back to grainwater. This is so much more 'see through' and colonizes just as fast. Btw, I have also never used stirring devices for LC

If there is still some undissolved LME floating around in the water after PC is that because it wasnt shaken enough before pc? Hope it's good.

It's about dissolving the nutes into the liquid with heat. If i want it really chunk free I'll also strain it through cheese cloth before PC but it isn't needed. The myc will eat it all. Chunks are fine.

thats not how you get clear clump free broth.
its also purposely written in the tek why you DONT use heat to 'dissolve' the nutes during the prep.
Quote:
add the powder first THEN add COLD water. this prevents clumping.
do NOT use hot water to make broths the hot water can cook the grain powder giving you lumps
nutes will be dissolved during the cycle but if you use hot water to prep then the powder wont dissolve.

then dont give advice on how to make it

You have to use a really low amount of LME or there will be sediment no matter what you do. I have 2 LC's going that are 2.5g LME + 20g dextrose and those are pretty clear. This might be my new recipe. But the sediment doesn't matter. Myc seems to like it. Your jars just won't be as clear.

I just made my first LC. I made mine practically the same as I've been making my agar media... With beer.
Looks pretty clear too. Hope this cambo LC does well enough to save me some time expanding. Thanks for this thread mushboy you rock

Haha, I know better.

I literally JUST inoculated it so no growth Beer has done me well in making agar culture media (or just "agar" as a normal person calls it) so I figure *fingers crossed* it should do well.

There is a difference between clear and clump free, in my opinion. I've made a ton of lc and that's how it works for me. I don't give a rats about clear LC but i don't like chunks and sediment. Heating the water on the stove after adding it to the powder is how i attain semi chunk/ sediment free LC or agar. To make it nice and sediment free I'll strain through cheese clothe.

Or follow my tek and use cold water.

Heating on the stove and cheese clothes? Hard pass.
