Nice write up! If I may propose another simple recepy. For those of us who regularly eat boiled potatoes: mix some of the potato water that is left after boiling with some regular tapwater to get very clear LC

So, most of these now aren't using stir plates... any reason why? I've noticed that with a stir plate I can get close to a full 200ml of usable LC within 3-5 days with a stir plate.
Also, I've been going dirty on my recipe. 200ml water + 2.5g LME + 1tsp (approx) dextrose. I get much faster results and the culture looks clear within about 3-5 days, but still brown and full of neutes.

How in gods name would you flame sterilize the whole loop enough to make it "clean" to go over the top? Seriously, not trying to be annoying, I'd rather do this than waste a whole syringe on testing it out.

Also use long ass loops.

I dont use stir bars because I dont need lc under a deadline.

Just checking in on my liquid and I found something perfect for the conversation. The first two pics are the same lc. It's been neglected for a while, full of chunks and has an entire myc layer on top. I'm pretty sure it's clean. I'll throw it in a bag and check.... In the name of science. The other pictures are random other LCS that are tested on agar and clean. One is already in a bunch of bags and the other is on Deck.

Honestly Verum, I'd feel pretty confidant with that mycelium island not being contam. I've let a couple go that ended up doing that, put them back on the stir plate and they went back under and turned out fine. My guess is that the myc prefers to not drown while it grows... this is very unscientific of me.
Though... when that happened it was a huge PITA to break up the myc and it got lodged in my syringe and... well, it eventually worked so Still, not contaminated.
I'm excited to see that grow out.

These were forming on top too but I swirled a few days ago. That layer is just suspended now. Very neglected but grew out clean :reckon:

As I was straining I had a thought....Shit, how about heavily salted potato water?? Can't boil potatoes without a shit ton of salt.

Also, good point on the salt. Have I ever commented that every time I make grains I have the strongest desire to add salt and pepper? Every. Damn. Time. I once had to stop myself, I had the damn box of salt in my hand!

Why do you only drop in a wedge? Not a huge chunk of an entire plate that's been cut up tic tac toe style?
Easier to break up/agitate or just less chance of contam being in a smaller wedge?

most folks advise putting a tiny piece of agar into an lc to drop the chances of transferring something other than cube myc. Nutritious Liquid is a god damn feeding frenzy for any living thing. Any contam at all will completely ruin the entire lc. The same agar may do perfectly fine in a grain jar. A lot of peoples agar is not as clean as they think it is.
Also, LC colonizes plenty quickly with a tiny piece.

So, for the vessel lid, any solid/regular vessel lid will work as long as it has a synthetic filter in it? (On account of the exploding and infection factors). Am I correct here? This is a rad tek for the LC itself, but the vessel construction/use still escapes me. Maybe I'm over thinking it.

- Popping Corn Rapid Re-hydration - Half fill all containers with corn. Empty into Instant Pot. Cover with water. Run 30 minutes on PC. Release, remove, dry.
- Wheat/Rye Berry Soak Re-hydration - Half fill all containers with grain. Empty in pot with water. Soak for 6-12 hours, air dry until surface is completely dry.
- Grain Jar Sterilization - Run 2 - 2.5 hrs on PC
- PastyPlates, MEA Formulation- Run 45 minutes on PC
- LME Liquid Culture - Run 45 minutes on PC

I dont put filters on my lids. Dont want to mod the media bottles cap.
Leave the lid loose and covered with foil. Just like making agar. Same exact process.
