Nah. But it is recommended that you put something sterile in there to break up the myc when shaking. Some people just use a piece of broken glass for example.
Stir plates are super-science-y fun though

Wow, this thread is all over the place right now. And i'm not tripping.
I actually have thought of extracting actives from LC. I even talked about with my super sciency nerd friend that is kind of a genius. He one upped my shit like a mofo though. And said, of course it's possible because.... Blah blah blah about chemistry shit. and then continued, "but why would we even bother, let's just program, I know you hippies don't like the word genetically modify so... Let's program some bacteria to shit psilocybin. It's totally possible. That's how the...." The dude is a trip.
I pour just a tad into grain jars and at least three or four tads into bags. I think a tad is the super scientific way of saying "just about as fast as you can pour without being a tweaker. Like a super stoned person trying to pour just a drip but accidentally getting twenty five drips or so.

And how much agar do you put in the LC? Let's say you're going with 350ml water and 1.5g LME?

The less you put in the longer it will take. I would recommend starting small when you're new to this, so probably like a 1/2" x 1/2" wedge or smaller. I usually use about 1/4 a plate for about 250ml. I know people who use a full plate of agar for 500ml... seems like at that point just do a LI rig

I just cut a little piece out like i'm making an A2A transfer. While it is true that more myc equals faster colonization of the LC, I do not find it to make a huge difference. Also worth noting, Nutritious liquids are virtual orgies for living organisms. Agar isn't always as clean as one might think, The less you put in the greater your chance of only transfering cube myc. Some recommend putting in the smallest amount possible for that very reason. I don't though, just a little standard size transfer for me, no matter the size of the LC. I generally make 1000 CCs of LC at a time, with a tiny little agar wedge.

Rhizomorphic mycelia With LMEA i do 2% agar to 500ml distilled water, pc, x4 1pnt jars fill 3/4with distilled water, topoff with LMEA to %100 1pnt,pc then let cool. Now you can use for your cultures.
Tomentose mycelia Use 1% agar to 500ml water, then follow rest of directions for 2%...
The piece of agar i use to drop into my LC, is as wide at a #2 pencil lead tip, and about 1/4" long, so by measuremeants it'd be roughly- .10"x.25"

I started using larger wedges for this because I was sometimes having trouble getting the small transfers I do for A2A to drop off the scalpel. Basically the little wedge would stick and not fall off.
I find that the difference between the "grain of rice" size to the 1/4 plate to be a significant difference in colonizing time. With a stir plate + 1/4 wedge~ish I can use the LC within about a 7-10 days. I was fucking around with half plates and I could get enough myc to use the LC in about 4-6 days. With the tiny transfers/josex poke it would take 3~ish weeks until I could use it.
Granted, this doesn't matter for some people.

Finally cleaned up an APE culture I have been waiting to use to make LC... It's not super grown out yet, but I am thinking about taking one slice from about 12 to 12... Seem alright?

imo let it grow a bit more. when i take wedges for LC i want a really sexy leading edge piece that has had a chance to grow.
also take smaller transfers that cut is huge

I like to shake the fuck out of it like you're gonna pop the bubbly after you win a NASCAR event. This breals up the early growth pretty well and then I swirl once I have lots of growth not just agar fuzz ball jazz.

Been running lc in media bottles without a filter. All those ksss tubs were done from lc using a media bottle. You really dont need that much ge i've noticed plus they work really well for pouring (duh)
