I knew I'd get told to test on agar...the thought of pouring that liquid onto a plate scares me. I'm for sure going to make the plate wayyy to wet. No? Am I missing something?

Make an inoculation loop, flame sterilize it, dip it into the lc then streak it to agar

I plan on making a new batch or 2 of LC and using media bottles and pour to inoculate like mushboy shows. Until then I have a jar with a whatman and a SHIP in the lid. I recently got a bunch of 10ml syringes with shitty slip needles too. I have regrets.
Would you think using the SHIP and tilting the jar to pull LC, one syringe after another is the best way to go? Or should I remove the lid and suck it out that way?
I feel like the SHIP is a vector but so is having the lid off long enough to fill 20 syringes, especially with that much hand movements. IDK it's like a pick your poison kinda thing and I just wish I had done it like the OP to begin with.

in the flow i can fill a bunch of needles without introducing a contam. But, your points are valid. Free pouring the lc is the easiest way to avoid these issues in my opinion

I just use an extra media bottle and pour it out directly as though I'm pouring plates. I figure if my plates come out clean my LC<G should too as long as my LC is actually clean. I have 19qt milo I inoculated the other day. Lost one to an accident ripping off a filter, and one jar looks off from the rest, but so far I see myc sans trich.

My girlfriend picked me up non widemouth quart jars a while ago and I havent used them because they look like a pain in the ass. I am going to use them for the jars I'm going to pour the LC into. I figure a bit smaller diameter mouth on the jar would be a tiny bit safer when pouring the LC.

I have my first jar going too. That’s good to know. I bought some premade plates and hope they are ok to use. Quick question... How much broth do you put on the plate?

Thanks Asura. I totally missed that. I’m a complete rookie to all of this and the forum members have been so kind and helpful.
Thanks for making me a success!

Well primelines a regular mouth jar does seem to pour out much better than a wide for sure. BUT I went ahead and simulated pouring out of a regular mouth quart jar that just had 250ml of water. I found that it has a tendency to run down the side a bit, which freaks me out. I also found that if you spin the jar around and pour out so the liquid pours out where the thread is nearest the top (nearest the lip) then the thread kinda acts like a ring of a media bottle and it minimizes the running down the side issue.

My bad, totally read that wrong. Got up and got baked this morning. I only figured that out for myself when I was weighing out my options for getting LC out of my regular mouth jar.
Fuck I can kick myself for not using a media bottle instead.
I just ordered 2 more just for doing LC and when they get here, ITS ON!

I've been using media bottles and probs won't go back. They work fine even with no GE hole. I agree with past though, those LCs look a bit cloudy.

Free poured about 10ml into each jar and shaken after pouring. Have a little leftover after the pours.
This is before the final shake n pours.
