What does it mean when a couple kernels of grain don't fully colonize?

Iv'e noticed this before, not sure if its the starts of some kind of sectoring.But ive noticed now
and again a couple grains wont fully
colonize. Is that normal or a sign of contamination?? Thanks for the help!

Well I can’t really tell you from experience.  My thought are that those grains are wet on the outside from a bloom of bacteria, so that prevented the fungus from colonizing those kernels.  So if you put that in with sterilized grains that might be a good medium for more of that bacteria to grow.  I’d like to hear another’s idea or experience on that.

It will become an issue. I've had similar jars that I tried to use as masters, those tubs we're so bacterial that they contaminated with mold before the first flush. Spawn them to shoe boxes is my suggestion. Forget about mastering them, you don't want to g2g anything that isn't perfectly clean ime.

To you I say meh. You still get fruits out of bacterial jars and imo if a noob wants fruits then it is completely fine to spawn them and reap the rewards. Suggesting for him to g2g would be terrible but throwing them in a shoebox at a 1:2 ratio will produce results. I've spawned way more bacterial jars and still netted an ounce and a half per shoebox after 2 flushes. Yes, it is something to look out for in the future, yes it is not optimal, but at the end of the day if it fruits, it fruits. If it doesn't, oh well, back to square one. Which is exactly where they would be if they just tossed it.

I am having a similar issue. I figure more pics can't hurt. What do you all think? It really just looks like grain pushed up to the glass and condensation, I have a bad feeling you'll tell me bacteria  :sad: This is 23 days of colonization. Condensation only appeared about a week ago, it seemed pretty dry for awhile.

I’ve spawned so much bacterial grain and It works.  I’ve got a shoebox that I spawned in March with far worse spawn that’s still putting out fruits, it’s out lived a younger shoebox that had better looking spawn.  So it’s fine.

Shut it down everyone. If your spawn isn't completely perfect don't spawn it. Who cares if you will get fruits and experience. Not necessary as a learning experience or anything.

Yes, at 30%. It took so long because I inoculated with a small agar piece and didn't shake it much after inoculation... This jar has been consolidating for about 5 days. I'm waiting for another jar to catch up before going to substrate. And, of course, maybe the potential bacteria inhibited :shrug:

How you would know if "Bacteria leads to mold" unless you actually did fruit a jar with bacteria? In fact, to become a good grower you would actually have to try fruiting a bacterial jar to have the experience necessary to become a trusted cultivator. And it seems that bacteria does not lead to mold in many cases, so you are spreading bad information (or maybe just incomplete information). Bad growers (or any hobby, profession, etc) don't learn from mistakes and experiment, this is what you are advocating.

I followed Bod's oat tek, agar tek, and everything to the T. You can see my Journal entry in my signature... I have had minimal contamination issues, but I have had them, so I am making mistakes here and there, but overall I'm doing all right.

All the jars have signs of this and it's unlikely the same contam came from my process in all 8 jars. Not impossible, but unlikely... I'm guessing the contam (if it is) came from the agar plates.  I only transferred once and the plates looked a little poofy in places... I was impatient. I have back up plates that I can clean up... I'm going to try to fruit and see what happens.

Cleaning plates up is the toughest thing when you're new. I'm still struggling with it at times, gaining a better eye for what's clean and what's what with time. Guess I should have just thrown all that spawn away instead of get the experience in though. Really fucked up there.

I like to wait until I see nice growth with no phantom spots. Growth that flattens out after the transfer. Too poofy shows me contams ime.

I know that bacteria leads to mold because I have 17 years of experience. I was a TC until I forgot my password.

There's no need to learn from mistakes that many, many others have already made.

Call me names all you want but I only advocate reliable methods, not spawning some half-assed bacterial crap. Making spawn isn't that hard. Use agar, do enough transfers to clean it up and spawn something clean instead of pulling some bullshit flush, thinking you did it right and never getting better.

That's all fine and good but it took me more than a few tries of bacterial spawn to get to somewhere I feel comfortable. Posting pics, learning from shared experience and personal experience, is imo integral to development in this hobby. I read for months before I even started and still didn't get a grasp until I worked further and harder personally. Maybe post some pics of what you consider nice and clean instead  :shrug:  :shrug:  :shrug:

No one said anything about never getting better. Fruiting from a slightly bacterial jar has nothing to do with not trying to prevent it from happening again.  Trying to fruit from this jar is not the mistake, the mistake was already made.

I've harvested over a pound dry from "bacterial spawn" in the last month. Some are on their 5th or 6th flush. Spawned to CCVG, yes, coffee too. Trichoderma city right? Wrong. I'll explain it as non sciency for you as I can.

Bacteria, the little nasty booger we mostly hate (there are beneficial species too, sterilize some straw and shit you'll get a tub full of green.) is more aggressive than cube mycelium and eats the grain faster. Cubes won't colonize rotten or spent grains but Trichoderma will.

Bacteria is at a distinct disadvantage because it lacks the mycelial network so it can't move across the grains. 9 times out of 10 those few uncolonized putrid rancid bacterial grains that are stuck to the jar stay stuck to the jar even after shaking it with a sawzall paint shaker adapter. In the event you do get some bacterial grains in the sub, no sweat. You only run into problems when large amounts of the grains won't colonize.

Trichoderma is a natural part of the process, it comes in when the mycelium is done to finish the job. It's the natural process. It becomes problematic when you just let it go and it adapts then you spread it to other tubs and it wrecks all your shit.

  TLDR - to be effective and efficient in this hobby it pays to have an understanding and respect for the entire kaleidoscope of organisms you are dealing with and the natural life cycle associated with each.

Rock on man. I don't distrust your experience, I just think we have a different view of the process. I'm here to learn, and spawning bacterial jars is a learning process. Anyway, I'm gonna put my jars to substrate and I'll update, good or bad. I'm actually a little pessimistic, but fuck it.

Don't be pessimistic about it at all. If you get fruits, excellent! If you dont, oh well, start a new project and learn from your past mistakes. Or just throw everything away and start over again. What do I know... Honestly, not much, enough to know that any experience is better than none.

G2G it then give the jar a smell test.  That will give you a clue. More than likely you will get fruits.  If it was me though I would have a backup going just in case.

I've never tried shoeboxes.  27 quart monotubs is the smallest I've done.

yup, my guess is that something is sneaking in with your agar or maybe your sterilization process.  when you say the jars looked fine for a week and then got 'wet' makes me think it could be endo-spores in the grain.do you have pics of the myc?  what times/psi do you sterilize at?

Here are the plates. Noob condensation prevented pictures of plates 1 and 2 from the top. Tomentose sections never sporulated, so I assume it's cube Myc. I knew the risk with these plates, but I through them in grain anyway. Plate three I just threw the very tiny sections on the left in the grain. The others I threw in quarter plates. I've been cleaning the plates in the meantime, so I got nice looking backups ready to go.

PCed grains for 2 hours at >15 PSI with the initial 10 minute vent. Condensation started basically right before full colonization.

just keep dropping wedges onto grain until u get clean looking spawn. try lots of new plates too and make some "blank" test jars  so u know your grain isn't the issue

Just to update anyone interested in the question on whether to fruit slightly bacterial jars (if mine were actually bacterial), the answer is yes. First flush this morning. Day 15 since grain to coir and this is multi-strain.

I’ll get bacteria so bad in bags, I’ve had slime In the corners, so I don’t use them all that much for grain any more.  But I have taken the whole block of grain spawn out and broke off the slimy bacterial corners and washed the whole chunk in the sink and have spawned that to a tub that went four flushes,  it didn’t trich until I after I dunked it.

I’ll put this here since it’s related.

I have not been able to get full colonization to oats. I’m assuming spore syringe to grain is just a bad technique.  I’ve recently started an agar to grain run so I will be able to conclude my theory soon.

I’m assuming these are bacterial. G2G transferred this batch 2 weeks ago. Shook 1 week ago at 25%+/-.  So far the myc runs look good until I shake. They recover quickly after the shake and then stall out and will stay in this half colonized state.

Have yet to see any contamination in my agar with the same syringe.

Excellent advice up in here. Glad to see people acknowledging wet grains as bacteria. It's up to the grower if he wants to spawn it. Sometimes it works sometimes it molds out. In a contam ridden environment, it'll definitely mold out, which is what alchemy was talking about. When there's a high spore load, you gotta have impeccable spawn, and all bacterial jars get the toss. Most noobs tho have a cleaner environment, and usually get success with bacterial jars. However as they see more molds and trich bomb their place, they gotta have impeccable spawn to get good results. It's for this reason, ideally subs get tossed before any molds show up.

That doesn't look "half colonized to me." It might be slightly bacterial. Compare to mine in this thread. The density looks similar, except mine has more bacterial nastiness. The fact that it recovers quickly means it's okay in my book. Spawn to a shoebox and find out. Update the thread when you get those sweet mushies :smile:

Just going to pop in and say that I don't consider minor bacteria to be a "tossable" offence. I spawn bacterial shit all the time and just put it in a box with a frowny face drawn on it. It has yet to affect most of my yields, even when letting them go out to 3-4 flushes, they do fine.

There is an unacceptable level of bacteria, when it's pooling yellow or preventing colonization in larger-than-part-of-a-grain radius then I avoid them. A little bacteria is kinda in everything :shrug: doesn't mean it's going to fuck mush up. If I was running a commercial mushroom farm I may be giving different advice.

I always tell people to spawn that shit while noting that it is bacterial so they know what it looks like. :shrug: Good work racoon, I would have expected as much if I spawned them jars. How many flushes are you getting?

I'm struggling to get the conditions correct with my second flush and it looks stalled. It has not contamed yet, but the conditions were atrocious. Wet and dry at the same time. They were pretty dry after harvest so I dunked 2 tubs (not long enough) and heavily misted the others. I kept having to mist the tubs because they would dry out. The surface was sopping wet, all the pins looked glistening for a few days, the condensation on the lids was non existent, and the sub looked dry after a few hours. Yesterday I decided to bottom soak them all (pouring water down the side and draining after a few hours). Now the conditions look pretty good. I live in the high desert, so it's pretty dry. I think I got lucky by accidentally going slightly over field capacity on my first flush. Dunking properly will probably be necessary for subsequent flushes in the future.

There's no flush tek :frown: After this debockle I think I got it figured out but it'd be nice to not have to fail to learn.

that sounds super dry.  try running a humidifier in there on a timer or something

So, here's how I do shoeboxes, but I live in the mid-west, so I have 40% RH most of the year. But I find that the best route for these is a set-and-forget route. If you're fussing with them more than once every 3-5 days, try a different approach.

What would you want to see in a "flush tek"? I'm still working on the above tek, and plan to include a soaking section for what to do between flushes. This is how long I soak, but people seem to think I'm doing it too long. :shrug: Screw it, this works for me.

So I shoeboxed the questionable bacterial / uncolonized jars at 1:1 and 2:1 coir trials. These jars looked very close to the picture above. I let these jars sit for about 6 weeks to confirm they had stop colonizing.

Here are a couple pictures of the shoeboxes. This is 10 days in I popped the lid to photo. They are currently at day 14 and no signs (that I know of) of pinning.  No  pseudo top layer but I will next time.



Is the grain stalling issue caused by bacterial or simply multi strain genetics via spore syringe? Does this translate to inability to pin or slow to pin?

My Current theory: Do multi strain genetics lead to mycelium stress, which causes jars to stall, which then promotes a bacterial environment?

This is where my inexperienced beginner deductive reasoning has led me.

How fast did you expect them to pin? 2 weeks is nothing, seriously. I don't see a stalling issue, these look fine. You'll see pins soon.

if theres bacteria in your jars it was either on the grain already  or it snuck in with your inoculant.
