Sorry if this is a stupid question but can't seem to find the answer. If made some agar plates and slants at the same time but didn't want to use the slants right away as I want to work out some clean samples on the plates first how long can I store the slants before I use them?

Pretty much indefinitely. But i would personally make new ones if i hadn't inoculated them within a month or two
|

Do you have to crack the lid of the media bottles before reheating in the microwave, or can you leave them sealed?

Thanks a lot this really helped mine is cooling as I type .. I think everything has gone well so far it took awhile for me to find a recipe for mea I know it’s simple for most but there seemed to be a lot of conflict on the topic I went with 8 grams agar 15 malt extract and 500 ml water if anyone is wondering.. thanks again

1-4% nutrients w/v% works great for most agar anything. I like to stick around 2% with malt

hey i just got one question: where do you buy your plastic petris? I can only find polystyrene ones on amazon, and as far as i know those can;t be pressure cooked.

Oops.... Wait wait wait. You can't cook any plastic petris. You aren't supposed to. You cook the agar and then pour it into the petris. They come sterile.

I just want to add this because I make heaps of agar (day job)
Instead of microwaving to boil in the schott bottle I stir the agar mixture solution into an open plastic jug. I chuck 2 jugs of 500mL (half a quart??? two cups?? sorry I don't know what that is in gallons) of agar in the microwave for 10 minutes. Stop every 2 minutes to stir, usually it will start to boil in the 8th or 9th minute when it does stop it and give it a good stir. Then put it back in the microwave and when it boils again take it and stir it one last time. By now it should be fully dissolved, transfer to your schott bottle and autoclave or PC as normal.
Only thing is I use pre-made agar mixtures. Results may vary with your own agar mixture but it will boil much quicker this way as the schott bottles are made from thick pyrex so heat takes a while to penetrate. Also the plastic jugs are 1 Liter (quart, 4 cups, 1/4 gallon??) to allow space for the bubbles to rise and not boil over out of the jug. This way if your late to take it out the microwave there is heaps of headspace so you have plenty of time before it boils over and makes a mess

thats what i was thinking. after reading all the agar teks i realized they werent forgetting to pressure cook them, it's just not necessary as long as the agar itself is sterile.

Where are you getting bulk plates? Even on Amazon the plastic are more expensive than what you’ve posted.
