If you have cube plates, it should move quicker than those.

Thanks. I appreciate the answer. I’m down to a last bit of spores that may be good may not. If I run into this again with that print I’ll have to do whatever it takes. So I may take you up on that. I forgot to add that I did transfer from those plates and has since exhibited the same thick fluff 😡 Using a stout lab mixed PDA though maybe too nutritious? I hardly get the really rhizomorphic mycelium with my cubes either. Though never as tomentose as my Pan culture is atm

Hey asura I recently spoke to you in a message. I'm the dude having difficulty isolating a good phenotype. Well the 4 transfers I took after speaking to you are all slow compared to EQ cube isos I took on the same day. (Took them to compare speed) temps are 77-80 and they're only half the size of the cube myc. something just occurred to me that I have been using antibiotic agar. 25mg gentamicin per 500ml MEA. Could this be the reason I'm getting slow growth on all my pan cyan plates? Edit: hoping speaking about this on this guide instead of by might even add some more good info

Don't know the solution. I use plain MEA for my pan plates. They don't all grow fast...but the good ones do
Finding a good one can take some time if you can find it at all. I have so many so so grows that have validated this for me. In fact, I just posted an example out on the pan thread. I fruited something I knew wasn't going to work, but I did it anyway just to get more prints.
You can start again from spores or try to fruit something that looks good (like in the pic in the write up). That's what I do.

If you could guess roughly how many plates does it take you to get promising growth? Those 4 transfers were just the last 4 lol I have a stack of these plates now. I'm waiting on some petris. When they arrive I'll start some fresh from spore as well as transfer some cultures from the antibiotic plates to regular MEA to see if the gent was making an impact. I transferred the quickest colonizing parts of multiple trays only to end up with slow mycelium. Oh well. Something will give eventually.

is this coming from your own frustrations trying to isolate one?

@Asura: Can it be that sometimes after a x number of transfers you get faster growth?
I noticed faster growth on the leading edge of my T6 dish and transferred that faster bit (@ about 02:00).
That look more like the 'whispy' growth you refer to?
Oh and btw: some pan pins are popping up again. Hopefully this time I don't screw them up. Pics will follow in the Pan thread

@Tweeq I've never gone past 5 plates. But yes, sometimes I get fast and wispy after a few transfers.
That Ogre Boy you sent never got the growth I was looking for after 5 transfers so I chucked it. I still have the other print so I will start from spores again when I have time.
Your plate as a whole is not would I would look for (uneven growth, the rings) but I am the first to admit that I just don't know. I like my certain phenotype and try for that above all else.

@Asura thanks for elaborating! Yes the plate looks weird. Started out with uniform growth, slow and cottony and the more orregular, faster growth is new to me. Took a T7 from the outer edge (the fastest bit) just out of curiosity. If that one grows uniformly I will make my next batch of LC out of it and we'll see how that goes.
This is my only Pan clone culture so I'm reluctant to chuck it. As a backup a ms T2 plate is waiting in the fridge and I've saved some spores of course.
Some pins in my fastest pan tray!

that was not clear in the OP maybe you should update that. Especially since this is one of the better threads on the subjects.

The bit on transfers? It's not unique to pans You can get different growth patterns thru transfers and/or switching media.
I mean the OP does say that if I don't get what I want I chuck it in the trash. Were you thinking something else?

Not sure how I missed this one but the syringe of pans I have had for awhile have been in the back of my mind lately. Good write up.
