Clone morels?

Since the stem is hollow (at least from the ones I found), how would one clone? Just scrape a piece of inner stem tissue with scalpel?

Only have have experience with cubes. Thanks advance.

i know it will be a PITA, however i will try to pick a tinny piece of innner tissue with a sharp brand new scalpel. scraping should also a be good idea i guess.

Do it! i want to see the results.  I would think they would be vigorous, I’ve had very little, but some, success just throwing my cutouts  and wash water around places.

What you can do is cut a section with a scalpel or try to tear a piece off with forceps and then dunk that piece in a freshly made 10% bleach solution for 1-2 minutes then transfer to your cloning media. The mycelium will survive this, but it will kill some surface contaminates. I did it this way and it worked, but I did have bacterial and fungal contamination in about 60% of the transfers. But some were relatively clean.

If you transfer to an agar media you can clean it up that way. Morel mycelium grows relatively fast and is on par with oysters for how fast it expands, so be prepared for this.

I just transferred a small Piece from the inside of the morel to agar last night. I was not sure how to do it either. When I picked the morel I basically dug up the whole thing so the stem was still sealed. I will update in a few days. Or once I see growth.

Here's 3 different plates of black morels that are 2-3 weeks old:







Sorry for the picture quality. The third one I'm unsure about. Dunno if the mycelium turns darker as it matures or it's mold. I think it's mycelium based off the 2nd though.

Yes, black morel mycelium, based on my limited experience will turn orange to brown as it matures. The sclerotia will also be an orangeish color, similar to the color of the inside of the cap.

I got nothing but bacteria on all 10 of my plates. Going to let them go and see if anything else grows and I can salvage it.

That blows. My half-free morel plates shared a similar fate. Blotches of trich all over it. There is minimal mycelium growth next to a sample between the trich I can maybe save but doubtful. It's going to be a very tiny wedge.

My black morels on the other hand developed rather nicely.



I transferred wedges to plates, some extra rice jars I had made up for cordyceps, and LC.

Newtomyc, what was your technique for cloning if you don't mind me asking?

I've been largely successful shaking the mushrooms in a solution of 10% bleach and water to wash away any surface bacteria. From there, they're placed in my SAB on a paper towel soaked in 70% iso alcohol. A sanitized razor is used to cut the specimens in half in any way possible and a flame sterilized exacto is then used to cut or scrape a piece of freshly exposed tissue for transfer.

EDIT: The aforementioned trich was not from the sample itself 😡

I did it like a cube. Cut open in front of my flowhood and cut a little bit of inside tissue out. I did wash the morel but just in a vinegar bath.

I will try your tek out. This was my very first try at anything other than cube. I have a huge batch of morels in the backyard. They have been coming up every time it rains.

I wouldn't recommend it, I misspoke. The exacto was flame sterilized. As far as I can remember, the black morels were my very first attempt at agar work too.

I found the half-free morels about a week later and had to figure out what was wrong with them because I had never seen a morel like that lol. I did some lion's mane and scraped some cubensis prints at the same time. One of those is 100% clean and the other has quite a few specks of black mold and they were done exact the same way within a minute of each other.

First batch of agar plates I poured I forgot to put nutrients in, wasted half my disposable plates lol.

Shit happens. I don't know that the technique that I used was good or bad, just made the most sense to me and I was trying to figure out why you were having issues with bacteria. I've seen black mold and trich, but nothing else. The bleach solution was just to wash away dirt and kill surface germs. Cutting inner unexposed tissue seems key to sucess.

The half-free morels I scraped a sample of mush because there wasn't inner tissue and it barely grew it all. I'll try to get a pic real quick.

EDIT: Nevermind, trich landed and grew off the sample. I cut a wedge prior (probably when it happened) but it didn't do anything on the new plate. Don't know if scraping what amounted to slime was to blame or not.

I wouldn't even bother with the bleach. Simply putting the smallest piece of inner tissue into a jar with sterilized water shake and then plate the washed tissue. This will preserve vitality and vigor compared to sanitizers as well remove contaminants.

Correct me if I'm wrong but bleach doesn't harm mycelium? That's why I chose it over peroxide.

Again, I'm putting the whole mushroom or sliced mushroom in the jar for the wash. The inner tissue never receives any contact with the chemical.

My line of thinking for using the razor and exacto is that the razor is just sanitized and used to cut the sample in half. If possible, I start the cut with it and tear the rest in half leaving a segment that never made contact with the razor blade. This is easier said than done of course working with something tiny and it may be overkill.

Misread some of RR's advice. He stated it wouldn't kill live mycelium, not that it wouldn't harm it. In this case it's tissue anyway.

That does explain the mystery of why my half-free morel tissue showed little to no growth.
