P. Cubensis Natal Super Strength Bulk Coir Cultivation

Hi all!

New member here having some trouble with a bulk grow. So I've grown a few times successfully in the past but haven't grown P. Cubensis since April 2017. I recently decided to start cultivating again and got my hands on a few strains,namely Cubensis B+, Natal Super Strength (NSS) and Albino Penis Envy. After I received the spores I made LC's and then inoculated WBS spawn jars.
The NSS seems to be a Natal/Transkei strain and finished colonizing the jars in about a week, the B+ had contams and the APE's took a bit longer. On the 14th I inoculated two large bulk tubs with the NSS. I inoculated at about 3:1 bulk sub to spawn ratio. My bulk sub is a 1:1 volumetric ratio of coir to verm respectively. I added a 1-1.5cm casing layer of 1:2 coir to verm at the same time as inoculation. I then incubated the the tubs for 5 days until the mycelium was about 25% colonized through the surface of the casing layer and I could clearly see it coming out everywhere else. I initiated fruiting conditions on the 19th April by exposing them to 99% humidity and frequent air exchanges via air pumps. I've made sure to maintain the temp between 21 to 25 degrees Celsius. After three days I noticed a lot of knotting happening on the surface and I assumed it was on the verge of pinning. It's been a while now and I haven't had any pins form. The knots on the surface have just been getting thicker and thicker and most of them are at least 1cm in size now. I have never seen this before because usually the mushrooms started pinning through the surface of the casing layer. Ive attached some pics and maybe you guys can help me figure out what on earth is happening here. I know the knots to be mycelium and not mold although the pics might not do it justice.


My camera glass on my phone has some dust behind it so it takes pretty blurry photos. I'll upload better ones tomorrow that I'll take with my friends camera.

The best way I can describe the balls is as solid knots of mycelium that are pretty fuzzy. Its a bit worrying that they're covering the entire surface. Will mushrooms grow from these or should I scrape the surface and refruit? Maybe flip the cakes and refruit that way? Let me know if you guys can deduce anything so far. Maybe I'm just being impatient, or perhaps the shroom gods are pissed at me for the sabbatical I took? They're my babies so it's been stressing me out a bit.

Put your spores on agar next time. Spores to LC is a gamble, and its doubtful your culture is axenic

3 weeks to fruit isnt unheard of. But those tubs look suspect to me like theres something else growing up in there too. The thick-milkshake looking growth is indicative of bacteria

1:1 coir verm is very heavy on the verm, should be more like 4:1.

Evaporation is a pinning trigger, don’t mist unless the surface dries out.

Be patient they’ll probably fruit. And instead of worrying about these tubs start workingnon agar so your next ones will be better.

Thanks for the response bud.

Yeah I should've done agar first but I was a little keen to get it going right away. So you rate it's the bacteria making the mycelium grow like that? Would that be surface bacteria or bacteria in the substrate? I thought it's maybe some strange structures the mycelium is forming from lack of FAE.

I'll definitely make some plates tomorrow and get the purification going for each strain so I can be 100% sure my LCs are axenic. I'll also adjust my verm to coir ratio for the next inoculation, too bad I inoculated the APE on Tuesday already else I wouldve adjusted the mix a bit.

Below are some better resolution pics so you can see more of what I'm dealing with. I was able to get pics of both tubs this time. They're inside a larger Fruiting Chamber which I mist and fan regularly. You'll see that the mycelium is also creeping up the sides of the tubs.

IME the most FAE you can give a culture without it drying out is the best.

I’m not 100% sure that it is bacteria but bacteria is everywhere and one the main signs its riding along in your culture really thick myc that looks like ice cream or a milkshake, it will form a snotty slime on grains or subs if theres enough liquid,. I think a cloudy LC is a sign of bacteria too but I’m not 100% on that I dont work with liquids very often.
More fae and less moisture can help the myc fight back,but it  competes with your mushrooms for food and the mushroom has to spend energy fighting so your yeilds will be lessened, and susecptibikity to other contamination is higher.

That's an odd looking sub. But I mean.. If it fruits it fruits. Let it be.. check daily to make sure their is not an aliens on it.

Okay so I've gone ahead and stopped the humidification for now and I'll let the tubs evaporate for a day or two. Would you guys suggest I scrape the overlay or leave as is? Has anyone ever seen this type of growth before?

As far as I could tell my LC's weren't cloudy at all. I used filtered Malt Extract broth as the growth medium for my LC's so there wasn't even much residue in the cultures, maybe a couple particles. But to be sure I'm going to get some selective plates and test the LCs for bacteria, I might do the same for the surface of the tubs.



I'll definitely make the effort to mate the strains a little bit better while I cultivate them on the agar. I just finished making some agar plates for purification so they're cooking in the incubator atm.

Also any suggestions on fruiting my APE's so that they don't have the same problem? I inoculated them with 30% spawn on the 23rd so they should be ready to start fruiting tomorrow. Will attach pics or make a separate thread once I check on them and start fruiting.

What kind of plates are you using that are selective for bacteria and not fungi? cycloheximide is restricted and not exactly dirt cheap.

The University near me has a Microbiology department that sells supplies on the side, they charge me .25c for agar plates and $1 for selective plates. I'm not sure what they specifically use for their selective media but I am placing an order now and I'll ask them.

I do. Yeah, bacterial LC is cloudy (maybe not always... I can't definitively prove that... but in my experience). If a teenage LC looks cloudy and has since you mixed it up and cooked it, don't trip. Give it time. The cloudiness is potentially just little bits of nutrients. As the Myc grows it'll eat up said tiny bits and the LC will clear up if it's clean. Even so, one should always test LC on agar before going to grain.

Using proper sterile tek in the flow or SAB, you dip a flamed loop into the liquid while being very careful and aware not to contam your lc in the process of checking it for contams. Streak this Loop across agar and you'll know in about three or four days if you've got problems.

LC made from spores is a horrible idea, in my opinion.

Your tub does look strange and i would guess that it's a little heavy in the bacterial department but i wouldn't say it's a SURE thing. I'm a little unclear on your fruiting situation but you definitely don't want to keep it too wet and some of your pics look a little wet. Let her breathe and re moisten if she dries out.

I've had some cultures do weird puffy shit like that and then do alright but i'm certainly not excited to see a tub doing that type of stuff.

Thanks a lot for the input guys. I figured its a combo of bacteria and too much moisture since I suspect water was condensing on my spray bottle and dripping into the tubs causing pooling. I've left the FAE on for a few days now without any misting and the humidity is still around 90%. I'll keep letting the pooled water evaporate but I suspect its quite a lot. If they dont pin in so 3 weeks time I'll flip the cakes and then see what happens. Hope to post pics of some pins soon tho!

Fantastic news everybody! We have pins!

I woke up this morning and gave the cakes a quick look-over when I noticed one of them had formed several little pins at various spaces all over the sub's surface:grin: I took some pics but they aren't showing up at all on the photos. I even had to go back and check to make sure my mind wasn't playing games on me. I'll upload pics in two days time once the pins are a bit more visible:grin: Thanks again for everybody's help, wouldn't have been able to do it without yall:)

The mushrooms growth/appearance will tell a lot more about the moisture content
