Pretty Myc Pics on Agar, Grain Time?

21 days ago I inoculated some agar with a B+ spore syringe. I transferred some choice selections to some new plates 11 days ago. Below are my transfers. Is it time to go to grain? And which plates would you use? The plates aren't as clean as I'd like (different morphologies and what not), but I think some of them look pretty good  :cookiemonster:

T1-C2, T1-C3, T1-C4. (Day 11 since transfer)


T1-C5, T1-C9, T1-E9 (Day 11 since transfer)


Note that I took some transfers from T1-C2 5 days ago but they aren't growing as vigorously as these plates. Also notice a new point of growth on T1-C2 around 3:00. It looks myc-ish, but how would it get there? Most likely a contam?

3,4,5 all look good to go to grain. If your transfers are moving slow its possible you introduced contams (bacteria) with your scalpel and thatโ€™ll be in the donor plate (c2) as well, either that or your agar was prepped differently/poorly/sugars carmelized from overcooking etc. Make sure to flame the scalpel red hot every time.

The satellite growth on 2 looks like bacteria which kinda backs up that theory.

Iโ€™d probably pick either 3 or four and transfer that fast sector from 5 to a new plate and both the nines need a little more work as well. Btw you dont need to wait for a plate to fully grow out before doing cleaning transfers, 4-5 days is plenty time.

Looks like mold from 8-10 oโ€™clock on #5&6.

Thanks. T1-C3 and T1-C4 will go to grain this weekend. The T1-C5 Rhizomorphic sections do look thick and great; I think I will transfer that one. And that's a good possibility on the next transfers, that satellite growth means I done fucked up. At least a little bit. It was a pretty disorganized, mistimed day in the SAB...

I might be, but I'm leaning towards its just tomentose myc. That being said, that stuff ain't going to grain cuz it don't look right. Those plates were actually transferred from tomentose/mold looking sections just to see what would happen. The fact that rhizomorphic growth came out of them on this transfer leads me to think it's not mold. However, I looked at those sections on the parent tray under the microscope and found some suspicious Sporangium looking things, but only a few and mold should have a lot more I think. It's also growing pretty slowly for mold... And it hasn't changed color...

I'd also say tomentose on pics 5&6. It's pretty common in my experience for the back side of the transfer to revert to tomentose, while the leading edge continues to grow out rhizomorphic. I'd take the whole rhizomorphic half of C9 and drop it on grain, or take some transfers from the right side, grow them out, wrap them up and throw them in the fridge for future use.

That's a good observation! I'll make sure to pay attention to that. And I think I will drop the rhizo C9 half in grain just to see what happens. And I'm optimistic about it.
