I've inoculated rye with a multispore syringe. I've always had a lot of bacteria and mold before so this time I thought I let it be for a bit longer in the sieve after I boiled them and stuff them in my containers and sterilize them when they are a bit drier.

I did it with 2 containers. One looked very promising at the beginning but now it has stalled and I haven't seen progress with the mycelium for a while. But it doesn't look really dry, I can see that there is some moister. I have even opened it and felt on some of the grains and they are a bit moist.

The other one is keep growing but the mycelium is much much paler than what I'm used to with grains and BRF cakes. Here is some pictures:

Is there a problem with these grains?

Also, I just learned that I shouldn't fill my containers like this. My containers are almost full. Could this be the problem?

What kind of contamination is this? And how can it be avoided next time?

I've never had success with grain spawns.

But the syringes works great with my BRF-cakes and they guarantee my that they are contam-free. Is that a lie?
I was always thought it's something wrong with my way of preparation and not the syringes.

Nah, it's the syringes.
Syringes are really only good for inoculating BRF cakes and inoculating agar.
When people inoculate grain jars with a spore syringe it's a toss up whether or not it's gonna get tossed out. Grain is not as forgiving as brf cakes.

I'm going to start with agar immediately. I didn't know I can inoculate agar with the syringes. I was waiting for my syringes to become empty before I start with agar work from mushrooms.

Yeah thats mold and bacteria. What container are you using? You sterilized in that? I guess, IF whatever you did to prepare that properly sterilized your grains, then yeah your syringe is probably dirty and youll need to do agar.

You should do agar anyway when doing grains.

I didn't click on those pictures before... What kind of filter do you have on those? Are you sterilizing in a pressure cooker?

Also if those containers are good enough and they might be, I wouldnt fill them up so full. When you eventually need to shake to redistribute grain it will be next to impossible. I would find some quart jars.

Those are very good. They have filter on the lid that keeps moister in but allows for air exchange. Made for this purpose.

I use a pressure cooker and I pressure cook on between 1.5 - 2 PSI for 90 minutes after reaching the PSI.

I will fill them with less next time. I will try half of it and inoculate with agar next time to see if I have better luck.

What kind of contamination is this? Yeast contamination?

1.5 - 2 psi???:confused:
Need 15 psi for about 2 hours.

Now I retract what I said previously lol. Syringes arent the culprit (tho spore syringe to grain is not good either), it's your sterilization procedure.

I'm sorry. I probably meant 15 to 20 PSI then. On my PC's screw, a scale is written out from 0-2. So max pressure is 2. I don't understand PC's that well but I'm thinking my PC can't be that shitty that it only offers 2 as max when you could need 20, or could it?

It actually could be your PC. Can you share a link or model #? I dont know much about the various PCs but I do know that there are plenty of "pressure cookers" out there that are utterly useless for myco work.

From the manual:

Working pressure: 0,6 bar (setting 1) and
1,0 bar (setting 2).
Steam releasing pressure: 260 kpa.

1 bar is 14.5 psi.
and 260 kpa is 37.71 psi.

But I don't know the difference between Working pressure and Steam releasing pressure.

I took my time to respond so maybe someone more knowledgeable could give their 2 cents but I hate to leave you hanging here, so I'll do my best.
From what I can gather yea 1 bar is equivalent to just a hair under 15 psi so your PC should be good enough for the job.
I THINK (emphasis on the word think) that steam releasing pressure must be like the point where the relief valve relieves pressure for safety.
But under normal operation it should maintain 14.5 psi
^ once again I THINK :smile:
If there is any kind of indicator for when it reaches 14.5psi (1 bar) I would set a timer for 2 hours and let it go.

I would make it priority to invest in a bigger PC like a presto if you can. Shit is way easy and you can fit quart jars. 

When you get into agar and you sterilize media bottles you will only need to sterilize for 25-30 minutes.

OK let's go
First you can sterilize at 0 PSI I'd do 120-140 jars every week at 2PSI
I think its more a bunch of stuff then just one thing.
The grains are wet
Contam that's from the syringe would show up around where the mycelium is
Contam that's from filter or jar not proper would show up on the top
Contam from not sterilizing properly would show up everywhere

Now you have some info to trouble shoot for your self.

On word
I think you should stick with PF tek for now
You'll learn much faster

Also learn agar as fast as possible!

You CAN sterilize at 0 psi if you run that shit for 8+ hours. Did you see the size of his PC? He won't be able to run that long without it running dry. Sure he could use a stock pot or something but why steam sterilize anyway, he has a PC.
His grain dont look wet either.:shrug:

Now his containers are a point of contention that's been addressed. Same with using a spore syringe on grains.

EDIT: I guess you slowly replenish water as you go, I had no clue before. I stand by it anyway, he has a PC, why not use it correctly. I'd hate to pay the bill from running 8+ hrs at a time, especially if ya make a habit of it.... I bet if you did that half a dozen times then your energy bill is going to be more expensive than a PC.
Hell if OP lives in an area where PCs are hard to find then I have an extra 16 quart PC that I'd part ways with if OP pays the shipping. I'll snap a pic of it when I get home.

I have not used the PC on max setting before. I thought the scale was showing psi so I had it on 1,5. Next time I will use max so that I'm almost on 15 psi and let it cook for 2 hours.

I will also prepare agar culture for it. I have already bought the supply for agar work. Scaple, Petri dishes and agar and all I need is to get some kind of nutrient for it. I'm going to try to inoculate it with both mss and mushrooms since I'm reading that people have been having problems with mss.

I live in Sweden so there are plenty of PC's I can buy or order. Thanks for the suggestion though :smile:
