opinions on my 3rd transfer plates please

before I go to grain in the next few days. this will be my first attempt at a2g oat tek, If I am not posting in the right place, please let me know, I am still unsure about this. TIA
Just looked at this again, I don't know where my brain was when I wrote the dates on them. I transfered the on 4/8/19.

Most look good but I would do another transfer and grow it out. I look for complete uniformity in my plates before I use them.

The second one on bottom from right, looks like you have some bacteria going on but it might be camera angle.

Thanks for the feedback, I was wondering if the round spot on number 8 as well as a few of the others we here there are what looks like 2 strains going on. If I use them I will cut away from those parts. I have about 8 more plates that look fairly uniform so I didn't take pics of those my plan is to cut the best looking sections tomorrow and drop into my oaks which I will start cooking drying and ping later today. I plan on cooking 2 sets of 10 jars, I think I should have more than enough plates for 20 jars. Still not quite sure how much of each plate should go into a jar. Quarter to half?

I use like half a plate cut into several pieces.

On your oat prep, make sure those are dry. The drier the better!!

If you wait until you have complete uniformity you're talking about isolating strains, which is gonna take many more transfers and then you're gonna have to test each one.

They're fine as-is IMO.

I am cooking drying and pcing today, I have them spread out in a sunny warm sunroom for 2.5 hours, they seem very dry. Thanks for advice everyone

That's not true. My first plates I grew out were completely uniformed in 4 transfers. I'm not isolating strains, I'm just making sure I have clean cultures.

MEA, 10 g malt extract,  10 g agar agar, 500ml tap water. I'm a total noob, so I'm pretty happy with how things are progressing.

Seems like your talking about a monoculture or "single sector isolate". Jst an isolate wud by a single strain, and that shit wud be difficult to get to.

As i undertand it, a monoculture is a narrowed down group of strains whose genetics all "get along" really well.

None of your plates look like a monoculture, but kinda close. Looks like when you transfer, you gotta roll/scoot the wedge around to the right spot and it leaves a bit growing in different directions. Or there is more than one wedge on a single plate, causing genetics to mix all weird.

I do the same shit. Lotsa times i get a contam spore growing on the edge of a plate cuz i smacked it with my scalpel trying to get the damn wedge off.

keep in mind that as you narrow down genetics you narrow down the possibility of the type of harvest you will get, if at all. Shudnt be worried about a monoculture i dnt thnk, but i wud test that shit before running a shitload.

Like I say, I'm a noob, my transfers are pretty sloppy, having a hard time getting the wedge of my blade, that's why the one is way off to the edge. But very likely there are a couple strains going on on a number of plates, not too concerned.  This is all from MS syringe and not from a clone, so who knows,  I'll grow some out, and take some tissue off a good one for my next go around. It's all just fun and games and learning for me. And again thank you to everyone for there generosity of knowledge.

If it's not an isolate then it isn't actually uniform. It may form a nice circle, but some strains just go faster than others.

Timbo, a monoculture isn't multiple strains who get along well. It is a single isolated strain, hence the prefix mono which means one of something.

To avoid getting agar stuck to your scalpel try to scoop it instead of stab it, then you can just sort of cut into the receiving plate and it comes off really easily.

Oh yeah that is a good idea.

Im pretty sure a single isolated strain wud be if from all them little hyphae you isolated one. However the monoculture may have a different deffinition. I do know that it's fun to play around with. Thats wat im up to now :cool:

They do look nice actually. Our styles are different. I take the smallest wedge i can. Yours are massive haha

You shud take a transfer from 12 and cross it with some of that rhyzo growth on one of the other plates. No reason why, other than it wud be cool.

I cant get my B+ to rhyzo. The first plate from spore did, and that's what i took my transfers from. Now everything is tomentose. Possibly cuz i use a much more nutrient dense mix.

Yeah I'm still working on my technique.
once i transfer from these plates i will wait until i get a healthy fruit to clone tissue, then play around. i  have some albino spores coming that i will put to agar soon, i took some grain juice that i will mix with my agar next and see how that works out.

Thanks for posting, I'm in the same boat... fresh to agar and doing oats! So I'm learning right along side with you.

Ive also been taking as small of transfers as I can... and figured out scooping after fumbling with so many stabbed pieces.

I cut a triangle. Start at top and go down and left. Go back to starting point and go down and right. For the bottom, I (try to!) cut at a slight angle from left to right making the triangle complete. Instead of pulling out my scalpel at completion... I just move it slightly back to the left as I very gently lift up.

Usually comes out smooth, and as was mentioned falls easily off into the receiving plate.
