Yea lol.

Skepticism is healthy, cynicism.. well that's your own problem ;p

That said, I was kinda skeptical about identifying monokaryons with the naked eye. I get if checking with a scope is not exactly convenient but it does seem like quite a good idea if you're not playing around. Might save a lot of pointless work.

The old agar pins shouldn't necessarily have to be so much of a problem. Especially since it's unlikely that they will putrefy in vitro.

But it actually brings up an interesting point: are the fruit's cells at some point all programmed for apoptosis (to stop wasting resources I guess)? Cause that would indeed be problematic for cloning.

I have doubts that the pointy caps were due to more than environmental factors, but it's a lot of fun.

Where are your gloves? Why are you touching sterile things with your bare hands?

What is the point in all this?

:horrified:

Why even use a hood if you’re gonna put your hands in the airstream.

Why name a P. cubensis “ liberty cap” which is a common name for P. semilanceata

Never seen a white mushroom with rust spores? what about the ones in the pinned post at the top of the page called rustywhyte ::shrug:

Irradiated spores?

Whats the logic behind storing petri dishes upside down? =

(serious question)

edit:

also what aye said. It seems utterly retarded to name a cube variety after liberty caps, the common name for a different species

I'm so confused by much of this thread. Maybe I can make better sense of it tomorrow but I doubt it.

EDIT: I apologize OP for dogpiling negativity although I think I might have been the least negative, or I'm fooling myself. The confusion was real tho. I might have posted the straw that broke the camel's back and I hate to think I ruined someone's day. I regret that and I'm truly sorry.:frown:

That's because they are RW with a look what I did bullshit story put in front. Isolated mono growth by eye and irradiated spores to mix moby dick and CRS. No one fell for that 😂

The spores I started with are all recessive for its mushroom and spore color.

All traits im aiming for are magically all recessive. Because I've heard that word before.

HAHHAHAHAHAHA Bodhisatta you're right I did hear that word before... wait I'm a Master Student in genetics  but thanks for the heads up, will let you know if the mono's came back true mono or not, btw the dikaryotic dishes (Which fruited) were from the undiluted plate, which were reasonably expected to be dikaryotic :heythereyou:

But I'm done with the negativity thank you guys for the nice atmosphere here :skol:

If you're a master in genetics you would know you need to form an inheritance tree to figure out if a trait is recessive or dominant or something else like sex linked. Which you've shown no pedigree analysis to prove this.

For instance red spores and leustic might be autosomal dominant phenotypes. People who don't really know what they're talking about with genetics 100% of the time will attribute a "bad" phenotype to being a recessive trait.

Quote:

In autosomal dominant disorders, the normal allele is recessive and the abnormal allele is dominant. It might seem paradoxical that a rare disorder can be dominant, but remember that dominance and recessiveness are simply reflections of how alleles act and are not defined in terms of predominance in the population. An example of a rare autosomal dominant phenotype is achondroplasia, a type of dwarfism. In this case, people with normal stature are genotypically d/d, and the dwarf phenotype in principle could be D/d or D/D. However, it is believed that in D/D individuals the two “doses” of the D allele produce such a severe effect that this genotype is lethal. If true, all achondroplastics are heterozygotes.

So can you tell me how you proved these phenotypes you desired are recessive?

Remember on this website most of your audience actually knows a thing or two so you need to show your math for ridiculous claims.

I know this because of the crosses which were made for Mister X by Una:

L = lijn (geen groei) = line (no growth)
B= barriere (dikke groei)  =  barrier (Thick growth)
O= Mycelium groeit om elkaar heen = mycelium grows around each other
G= geslaagd = succeeded

Because it was a dihybrid cross and the offspring showed a segregation of 9:3:3:1 (See picture above) and the 1/16 of this cross was indeed leucistic and has rusty spores

It's still immediately starting of labeling black spores and brown caps as dominant. They might very well be recessive.

Moby dick at the top left should be SScc but it's label is ssCC which indicates a rust spored brown cap

You’re right with the picture, I also told Una that there was mistake in that picture
But I added it as a kind of background information of the strain
