Flame scalpel after every cut?

Is it necessary to flame scalpel after every cut. I was thinking of setting up a small glass in my SAB filled with ISO to use to dunk and/or hold my scalpel. Just thinking it would avoid the risk of introducing new contaminants by taking scalpel out into the open air to flame it.

Alcohol doesn’t sterilize. Heating your scalpel to red hot does.
You don’t have to flame after every cut. If you cut a wedge from agar, you can immediately cut another wedge from the same plate. You just have to flame in between moving the scalpel from one item to another. Don’t cut the wedge, drop it, and cut another without flaming before moving to the second wedge.

Which in practice means you will need to flame every time I suppose. I always like to hit needles or scalpel with alco after flaming to cool them down too. Just thought this would eliminate need to flame as agar and jar contents etc are supposed to be sterile too. Oh well thanks for the advice

do not use alcohol on your flame sterilized tools. doing so negates the sterilization. you are making your blade dirty again by washing it with alcohol. you do not need to cool it down. just use it red hot and move on.

What munch said^. It only takes a microscopic bitch or two (that you can barely see with a microscope) to ruin your whole project. Flame STERILIZE, don't sanitize with alcohol. Alcohol has it's place, but not here.

Small glass of iso outside your sab. Dip the scalpel before flaming it to wash off agar residue so it doesn't burn on the blade

Noted thanks all. A small tub outside filled with alco and a few cotton swabs is useful too for wiping down jars before sending them into bat. Although from reading up on SAB it's more about trapping undesirables on surfaces so it won't fly around the SAB so soapy water might just work as well?

Great advice, and this is what I did as well. Sometimes reading just doesn't put things into full perspective for me. Bod's vids really helped me tremendously. I always flame after making each transfer whether it be A2A or A2G. Sequence would be: Dip scalpel in 90% ISO, then flame sterilize until red hot, then cool scalpel in agar prior to cutting out a piece of myc for transfer, then repeat until finished. The same method also used for inoculation loop processes for germination of spores to agar.

Personally I do one blade per plate and don't flame anything.

Yea I must get a look at those vids. If a pic paints a thousand words wat does a vid do?
Y'see when you take scalpel out to flame it do you set it back in the SAB and redo ur gloves with ISO?
