Peroxide Concentration in Agar

What concentration of H2O2 are you folks using for peroxided agar ?
I tried 0,3 %, but that seems to work TOO good. No contams, but after more than 1 week no noticable mycelium expansion.

10 ml of the 3% peroxide per liter of agar.
This means the final concentration is 0,03 %

And I'd like to add that I've used 5ml-15ml of .3% h2o2 in a liter of agar with success.

Thats 0.003% final concentration. I used 3 ml of 15% for 150ml of agar.

Ok,and I buy normally pre sterilized agar plates so :
How should I add H2O2 to the agar plate?
Prepare a mix with water and spray;does it work?I don't want to much extra water in the plate so :confused:

Unbelieveable. I was just preparing some new agar with lower concentration and was taking a last look at my cultures on the KILLER 0.3% Agar to choose which ones to transfer to the new agar with lower concentration - and there it was. I was trying to clone a store-bought Pleurotus ostreatus on two dishes, and on one of them there seems to be some kind of growth. Looks kinda weird currently but I will keep an eye on it.

Peroxide breaks down in light, so even if you use too much, it might
break up, allowing mycelium to colonise.

Stara, you can only add h2o2 while making up the agar mix, not
after the plates have been poured.

Higher concentrations of H202 won't kill the mycelium, it just takes longer for it to adapt to the media.

Good point Loki. Everything breaks down in light.
My inncubator is lightproof though. And the strangest thing of all is, that the cloning attempt started only 3 or 4 days ago. Must have been a fucking healthy mushroom.

About the presterilized dishes. Wouldnt it be possible to have a thin layer of liquid peroxide-solution on the agar. Although I dont know how much the mycelium will like the liquid layer. And the dishes would be damn hard to handle too.
