You're using way too much agar you only need maybe an 1/8"


Also buy some micropore tape at Wally world or order it online for your holes

I removed cap cut stem open and took tissue from inside of the stem. All done in a SAB. Took 2 samples from one and one from another. These are really shitty ziplock lids they barely even stay on like literally can’t pick up by the lid. As for the Polyfil I literally had no money at all and the poly fill around. And that sucks I had extra dishes to split that into. Will the poly work? And the recipe was 250ml popcorn soak water 250ml tap and 10g agar. Sound right? Also how long usually until the first transfer?

use light like this and you can see better those square ones are hard to see in I bought 40 and dont like them .  They stack better but visibility sucks and so do the lids .. usually with clones you should have growth within a few days I'd wait till you have growth about the size of a nickel ...


If you see bacteria you can do a warm pour or a sandwich .. to save the clone

My agar is so murky since it’s half grain water 😂. Plus the lids are blue so i kinda have to just guess when their ready. What is a warm pour/sandwich?

A warm pour is pc'ing agar then before it congeals pour it while it's still warm/hot over the bacterial tissue... a sandwich is when you cut a chunk of agar out and place over the bacterial tissue... the myc will outrun the bacteria leading to clean myc again..


You can buy different size wild bird seed at various places ... but all grains need to be soaked

I’ll try birdseed maybe. What would cause the slime looking stuff growing on the entire culture on my dishes? The tissue on top. Also the tissue blued hella hard.

Well the tissue itself will bruise and look a little slimy I suppose if on the agar there is no slime looking shit then it's fine.. but it also could be bacteria... when you make algae you wanna use food coloring to try to help detect nasties .... give it a few days and take another pic

Will do it’s only on the tissue so it should be fine as I’ll use food coloring next time forsure. And I’ll get at you in a couple days and let you know how everything is going.
