Good evening everyone.

I ordered these Synthetic Filter Discs online and they just came today.  They are a lot softer than the ones I ordered a couple years ago.  As you can see, I can easily bend them, they feel more like felt to me.  You guys think these are ok or should I try to find the more rigid ones I got last time?  Thanks.

Yes PCing syringes should be standard, but I always see people saying to fill with boiling water repeatedly.  So I assumed most don't simply PC them. It's so easy to do and you can sterilize a hundred at a time this way. 

I haven't tried metal lids because I figured they would be hard to drill and attach SHIPs and filters to

I've seen where whatman filters soak up LC and wick it to their upper surface and mold has grown on them.  When I said SFD I meant the SFD contained inside the whatman filter. If that filter gets wet it wicks nutrient rich liquid into it and is a vector.  The tube with lightly stuffed poly prevents that from happening. 

I use 60mL per bag.  I could probably get away with far less than this. 

I inoculate the LC by dropping an agar wedge into the jar in front of flow hood.  Then shake every few days. 

No photos of contaminated jars but I have these two which are ready to be used. 







If I leave them alone for about 2 days the mycelium will start to visibly grow along the glass.  It's very easy to recognize as it looks just like the leading edge of growth on an agar plate.  If I leave it for a week, the surface will start to colonize completely with a thick mat of mycelium.  I usually shake it or put in the fridge to keep that mat from forming since it will block the liquid from entering the syringe.

I think I will make a tek from this eventually.  I'll prep a few jars and contam them intentionally and that'll help demonstrate how the contams show up easily.

I wonder what those would look like if you stamped it down with a hot iron?

Oh yeah, that's right. I used to do that and I've recommended folks to as well. My current routine involves way more syringes, tweezers, and spare lids now though.

For me it's the opposite. I've always had issues getting RTV to stick to plastics, even with making cuts and sanding. So I usually put MP tape around the edges of the RTV holding SFDs on.

Ha, guess I'm a dumbass. I didn't know the Whatman filter was like that. I thought the ability to shake it and break up the colony without worry of spilling or getting a filter wet a major benefit? I guess it's because it's still so much smaller than whatever hole I'd put under an SFD. Your tube sounds cool. Sounds similar in purpose to this.

My buddy verum subsequentis is very solid with LC and I believe he does 40cc per bag. I'm hoping he sees your methods since it's pretty cool. I'd love to see a purposely contaminated one next to a regular one.

Wow yea those look a lot like felt. I actually used to use a double layer of "Craft Felt" from like a Micheals. I stopped using it though. I only use SFD's or polyfil pulled up thru one piece autoclavable plastic lids.



I got these from Fungi Perfecti, they're big SFD's to fit under lids for gallon spawn jars. They really don't bend much, they stay fitted up underneath the plastic lids tightly, it's hard to get them out.

I'm sure the ones you have will work but if it were me I'd look for the ones like these, they're inexpensive and will last forever.

does anyone here not wipe down the inside of their tubs with alcohol before you spawn tubs? if so, how is your success? i don't know why but i've always used alcohol. i'm thinking about skipping that step because it's a pain in the ass and sounds unnecessary...

fairly certain its totally unnecessary but I still do it most of the time. those old needless cleanliness habits in this hobby are hard to quit

i always clean my totes out with shower sprayer and use a wet iso rag before spawning.

yet i wont spray my sab down...:rolleyes:

was getting fond of bleach for cleaning SAB occasionally but didn't use it on tubs due to how clingy/hard to wash off bleach can be.

But now that I think on it, people use bleach/lime to pasteurize straw so trace amounts must not matter at all

well i always wash and scrub my tubs with soap and water after i empty them. but i have some extra spawn sitting around and if i don't use it now it's going to go bad. the only problem is that i'm really lazy. so i'm going to try a lazy tub run tonight and see how it goes. :shrug:

Yeah I dont think the bleach can do much harm I use like maybe 4 caps to a liter of water I mean I run into mold I'd hate to think it's been from me trying to prevent mold haha

:lol: when I was a newb I sprayed 20% bleach solution all over my bathroom before I did some SAB work. I sprayed it on the walls, floor, mirror, ceiling, sink, toilet, shower, everywhere. It left a nasty white film ALL OVER my bathroom.

If you use a 1:5 vinegar and h20 solution it will remove the white film left from bleach.
