Any portion of it... I hold the plate in my right hand,turned facing down at a 45Β°angle, scalpel in my left with the handle against my palm, and the blade run out between my fingers, pick up the 2 pieces of taped lid/ring action duo with my left hand,and spatula 8 of 16 "squares" (3 vertical and 3 horizontal cuts) into each of 2 jars per plate, using the scalpel turned on it's side...

I'm unsure if you mean inoculating the LC broth or inoculating grains or whatever with the resulting LC. I'd wait until the liquid is room temperature before I inoculate an LC. How long I wait until I use it depends on a lot of shit, e.g., volume of liquid, whether I'm aspirating into syringes or pouring, what I'm trying to inoculate, etc.

I've only used LC a few times, though, and have strictly done LI. While the majority of my LC's have been successful, I'm very far from an expert on them. One thing I've done on small-scale and planned to experiment more with are single shot LC's in baby food jars. The minimum time there would be a thick colony throughout the media. They make for easier aspirating and you can dump them into bags very easily.



This was a sterilized bag of oats + CVG inoculated with an LC in a baby food jar in my SAB. I have a bunch of little uses for BFJ's, including slants, regular plates, etc. Plan on writing a Tek on them when I have more free time.

Also I think it was you asking for it, but I did record me doing the Josex pokes. Or maybe you wanted Pasty's EZ LC. I believe I recorded that as well. I'll get them up one of these days.

I made up some lc a week ago and never inoculated it. Is it still good?

If y'all don't mind me slipping in a few questions real quick, that'd be great!. When is a flush considered done on pf tek? One of the cakes has one mush that is about 4 days old, and then a bunch of newer pins coming. Is that first mush considered a flush? Or do I wait to stop seeing pins before the flush is done? Second dilemma is, why the thin mycelium stretching out? to the left of this pic:

In theory, it should remain sterile, but how confident would you be that nothing got through? It's up to you. Liquids are capable of inoculating lots of grain and don't reveal contams as easy as other substrates so the importance of cleanliness is stressed. It's a teaspoon of malt w/ water and 45m in the PC to get a new, clean batch, so why not just do that?

I only shot that for folks requesting videos/visual aid on techniques. I have only went A2G a handful of times and have almost exclusively used LI > G2G since I started. There are many ways to skin a cat, but I couldn't tell you the optimal wedge size with any reliable data of my own, so keep that in mind. Fortunately it's easy to make plates and test stuff yourself and know for sure. :smile:

How many wedges, size, placement, shaking initially or not, softness of agar, etc. All things that could be tested out by folks.

I made some liquid culture and let it sit for 2 weeks.  It got a contam after this time and what I learned was not to trust my wattman filters so much, and now I keep tin foil over the top of all LC jars.  In theory, if everything is kept sterile, a LC should be good to use for a long time, probably at least a year.  As with all foods, time, temperature, and oxygen will degrade the quality, so it will eventually become less than ideal but who knows how long that would take.  It would take a long time. 

I use LC exclusively.

I use polyfil stuffed lids flipped for easier opening with no issues... seems i stuff a much smaller amount than most, ball about half the size of a ping pong ball, slightly pushed from one side, and then pulled into place really tightly...

Mr. Pig, how long you been sober? Pushing 3 years myself: quit smoking cigarettes Sunday morning around 3:30 too! Feels good,  congrats to you for staying sober brother man!
