LME and Agar advice.

I just got a half lb of light malt extract a brand new 500ml media bottle and a bag of powdered agar.  I was wondering what measurements some of you guys use to make the agar for the 500ml bottle?

My brain is drawing an absolute blank right now.
I wanna say a good starting place is would be 10g MEA to 10g agar per 500ml?

The agar that I use says to mix 1 cup of water to 1/4 cup agar agar, now my agar also has sugar added into so I'm sure that effects my ratios

Standard recipe
500ml water,
10g agar,
6g lme,

Germination recipe
500ml water
7g agar
6g lme

I personally would do 10g agar, 8g LME, and 500ml water.

If ur pouring softer plates to germinate spores to 8g agar, 8g LME, and 500ml.

well... shit i only have 1 media bottle atm.... and i have some clones and transfers to do.maybe i go in the middle?

Use the regular recipe for clones.

And u can make one regular batch and pour it, then pour a soft batch and pour them. Wrap the plates with cling wrap so they don't dry out and use them as u need them.

Well I should ask this first, what are u innoculating the agar with? Syringe or print? If ur using a syringe I would just use the standard recipe for everything. If not, split the recipe.

I have a few t2's on agar already, ready for t3 and i have a few prints of Rusty whites and PES Hawaiian im gonna use a loop on, all in my sab.

Oh ok nice. I mean it all depends honestly. If u have fresh spores they will usually germinate on regular strength agar. I use softer agar with dry old spore prints. And since they're rusty, I think it's safe to say they're fairly fresh.

I would just make regular strength agar. If it doesn't germinate after a week u can put a DROP of water where u put the spores, but I doubt u'll need to.

just realized i actually have 40 coming tomorrow lol... sorry a little stoned, so maybe i do 500ml of soft pour the 20 then do 500ml standard and label them.

do the 20 soft with spores from the rusty and the pes i just printed and use the other 20 for clones and transfers.

Umm are you sure, I assume you mixed your ingredients properly? How long did you wait for your plates to settle?

8g of agar should of been enough

Did u throw it out? U could put it back in a jar and add some more agar and resterilize.

What type of agar are u using? 8g it should have hardened...
