storing culture ideas?

hey folks..

just want to know where your mother culture slants are stored right now..

i have got some gourmet and medicinal mother cultures right now. I know there are many theories/practices of storing cultures for longer time. I am curious how you guys are storing them practically for longer run.

I too want to store them safely and keep my collection intact. any advice from those who are storing cultures for long time is greatly appreciated..

In the fridge. or in liquid nitrogen if you have that laying around :wink:

Just do some research for each species. Some species that are warmer weather will die in the fridge.

I know of no species that will be hurt by refrigerator temps. I store all my cultures in a freezerless mini fridge. It’s a mycology dedicated fridge, so it’s only opened once in a great while.

Freezerless mini fridge? I too want to have a fridge dedicated only to my mycology purpose. Could you please give me an idea of its specifications like temperature etc.

That’s an informative thread though I do put a stick in the slants to maximise their life. Just don’t know what temperature should I store them after they are fully colonized.

Why are the pinks an exception. At What temperature do they need to be stored ?

Which section/part of the regular home refrigerator do you store them .. the one close to freezer or the farthest vagetable section?

Im guessing around 40°. :lol: The wife allowed little area, I've got one door compartment with all of my stuff in it. Pinks are tropical ,cold can kill them.

I have a pink culture on order. Looking forward to playing with it for the first time. How do you store your pink masters?

I've read the classic distilled/sterilized water with a chunk of agar from some older threads. Has anyone found a better way? Seems like you'd have less material to pull from since it isnt going to spread right?

I thought of dedicating a mini fridge on a temp controller and heard 46f+ will be good for pinks/goldens but that seems like a bit much just for a few tropical species.

Easy , i dont keep a pink.:shrug: Golden seems fine in fridge. Although it has been a while.

i am too thinking of borrowing space in the kitchen fridge from the wife :laugh:
Planning to store them in a small tote or plastic box wherein i can store 20-25 slant tubes.

2 tubes dedicated to one culture .. are 2 tubes fine for each culture or i should have more considering to be on safer side.

That is all dependant on what you are doing. Im just hobby, I've got 2 - 50ml tubes for each culture. If my income depended on it, would probably have way more.

have read this and similar old threads many a times.. but why there is no post of dH2O storage of late.

also i dont think many shroomerites are using this technique thus no discussion about this here.

To be fair, not much talk in general. I know Gr0wer uses this technique. It's tested to work either way. Try it and post results.

Or better yet message some of the guys from those threads and pick their brains directly.

read this in a catalog of paddy straw :

The optimal temperature for
growth of V. volvacea ranges from
30-350C and the most suitable
temperature is 320C. Mycelium
does not grow at all when the
temperature is raised to 450C or

volvacea survive for the longest
period (Ahlawat, 2003).

Exactly, store at 15-20 C. That's just below room temp. Fridge is too cold for it.

For most oyster species I believe you can just store them in the fridge. I put my master slants inside of a gallon mason jar along with my LC syringes to keep any nasties in the fridge off of them. Mine have done well so far doing it like this. I just used one of the smaller round medium containers to make a larger master slant for my pulmonarius that way I can pull from it for a while, we'll see how that goes.

sop.. here's what i have got. 30ml Local anesthesia vials. i got them from my dentist friend who works in a government hospital and i had asked to store the empty ones a month back. wanna know how much he had stored for me ..? more than 300 :laugh: .. So i brought some 50 to make my Dh2o culture storage. the mouth of these vials is 1.5 cm. these vials has got self healing injection ports. once sterilized and inoculated i am planning to seal them with saran warp after putting the injection ports tightly.though i know that the injection port are good to last for many many years, but still..



what i am planning is to cut some 20, 0.5 cm wedges for each vial.  planning to dedicate 3 vials for each culture, since my collection has reached 13 (gourmet and medicinals mushrooms) :rockon:

you can also see one amber colored vial as well, he have some 50-70 of them. i chose the see through bottle so that i  monitor them time to time. my instinct says the amber colored are much better than the clear one, though i brought only one..

any advice is much appreciated especially from the old heads..

Maybe I'm misunderstanding, but you're planing on throwing 20 wedges into each vial? Why not Just pour some agar in each one and then throw in a wedge? You could even throw in a piece of wood and make a pseudo slant.
