I boil first because i've had it do that to me a few times when not boiled. I don't know the exact answer but i think that you should be able to do it without boiling first if you are pouring and swirl the bottle well before pouring. I just don't like swirling because i don't like bubbles in my plates.

yes, you should wrap them. Then store them on a shelf at room temperature. If you want to be really safe, you can throw them all in ziplock bags. I don't, but you might want to until you learn to wrap them correctly. Semi steady temperature is preferred as temperature fluctuations create condensation. depending on how liquid your plates are, it may or may not matter.

I don’t wrap until I use them.  Once cool, you can keep in a gallon ziplock until ready to use. Add dry ingreadients to media bottle (or whatever bottle),  top up with desired volume of room temp water, ensure no clumps on the bottom with a swirl or stir rod if needed and pc.

i don't use ziplocks but do wrap. it's easier and cheaper in my opinion.

How is it easier and cheaper to wrap plates before using, then unwrap to use and then wrap again after plating as opposed to placing 20 plates in a ziplock and pulling them out as needed?

I haven't done the math but i go through a lot of gallon ziplock bags. I may be wrong but it takes me seconds to wrap or unwrap a plate and one roll of cling wrap goes a long long way. i cut it into strips that are just a little wider than the edge of a plate. I don't like reusing ziplocks either unless i'm really careful to keep them clean. When i do sterile sessions i usually have my entire flow area stacked full of plates, jars, grain bags, syringes.... I have found myself being irritated with the bulkiness of 80 plates in gallon ziplocks.

old habits die hard though. i Just do it the way i do it because I've developed a process that works for me.

sitting here thinking about it though, you may be changing my mind as we "speak". I tried gallon bags when i was relatively new so it my might be time to revisit it. I'll be the first to admit that i am sometimes totally retarded and, as my father always said, "the only thing worse than being wrong is staying wrong".

Well I decided to just go forward with it. And hopefully I can pull off some good mycelium growth, and then transfer it to my second round of dishes. Just picked up 60 dishes online.

I decided to use a small segment of the Texas orange cap spore print I was given and used it to inoculate the 17 plates I made. Each plate is either bi or tri plate. So either two or three inoculation points. I used a pc sterilized pen knife to scrape the spores accumulating them on the knife. I then dipped that into a each Petri dish. Wiping with alcohol between each plate. Probably should have been between each segment of plate...

I then wrapped them with Suran wrap and placed them into a plastic bag.

you wiped your blade with iso between plates? uh... I hope the best for you but iso does not sterilize. Heat sterilizes. in theory your blade came out of the pc sterile, made the first plate, was wiped with iso and was contamed thereafter. I once did a test to prove this to myself and cleaned the everliving shite out of my gloved pointer finger with iso and then touched the center of an agar plate. bacteria grew from the finger print in every direction. You absolutely need a heat sterilization device for agar work. any flame will do but most use an oil lamp or butane torch.

So yesterday I washed and boiled some birdseed for spawn. I filled my jars and I had a lot left over. So I decided to try using oven bags as make shift spawn bags. (I do have some real bags being shipped)


I chopped the mouth ends off some pop bottles, ran the bag through the whole, then used polyfil as a filter to plug the bottle bag.

I did two runs in the pc. First run I took one bag with a quart of wbs and placed it on top of 7 court jars filled with wbs. Ran 90minutes at 15psi. Came out fine didn't melt, didn't burst.

Next batch I had three jars of wbs and a oven bag filled with a quart of wbs on the bottom. On top of that I placed a bag with maybe three quarts of wbs. I ran this cycle for 2hrs at 15psi.

They all survived the pc process. I did double bag the large bag afterwards.

Today I inoculated 6 jars and a small bag with a B+ Multispore syringe that was sent as a replacement for a bad syringe.  The other 4 jars and two bags were inoculated with a Texas Yellowcap multispore syringe that has been in the fridge for over a month. Hope all goes well.

How long will spore syringes last in the fridge? I also have live mycelium syringes how long will they last? They are edibles mushrooms if that matters?

I have 60 Petri dishes now so I'm going to do another Batch of agar. I wonder if I should let it harden in the bottle just so I know it will harden and I don't waste my Petri dishes.... I'll follow the same tek just stir like crazy and then shake the hell out of the bottle to mix it all together.

Can I use too much agar agar powder?
I want to use my live culture mycelium syringes on agar so I can save the strains.

so far I see nothing on my first batch of agar plates.

:smile: I stabbed myself with a spore syringe today... luckily it was unused and had just been flame sterilized.. ugh still glad I had that bottle of alcohol handy. Haha I guess mishaps happen.

Found out y my agar dishes never set... after reviewing my agar I noticed it was mixed with sugar. 10 g of sugar per serving. 15 servings in a 170g box. So I paid 8$ for 150g of sugar and 20 grams of agar powder :thumbdown: stupid mistake always read what your buying folks.

So when I get that I'll get my Petri dishes going.

A few days ago I Used 8 jars of colonized wbs to inoculate 4 shoeboxes and a monotub. 6 quarts of Roatan Honduras and two very slow to colonize B+ Quart jars. (3 weeks to colonize)

Today I finally decided I should try the BRF PF tek. So Im currently pcing 9 jelly jars
4 cups vermiculite
2 cups brown rice flour
1 Tsp gypsum 
2 cups water

I didn't think the gypsum could hurt so I added it as well. Pressure cooking at 15psi for an hour. And will hopefully have them inoculated tonight. Going to use some of the Honduras Roatan LC I've made. Also sterilizing some old spore syringes right now.

I'll keep everyone posted

So I've read that a few members like to spawn their tubs and immediately have them in fruiting conditions.  I however have not seen a post or tek showing the benefits of this.

I thought that mycelium liked the co2 buildup?


I have three shoeboxes that have been in spawning for four days. I'm thinking of putting one into fruiting conditions to see How it goes. (Dubtub)

do it. I've done a few tubs (shoes and monos) this way and haven't decided that they do better but have decided they do about the same. best tub i ever had was spawned and left with no lid all day every day. I put a lid on slightly askew at night. It need a lot of misting but preformed wonderfully.

Just moved it to a cooler room and added the dubtub. So do u case when u do this? Doesn't seem u would?

I pressure cooked a few quarts of peat moss, and vermiculite to play around with casing. I might as well play with the other dub tub.

So three 6quart tubs 1 quart of spawn.
Honduras Roatan strain
Spawned 1/31

Placed 1 into fruiting conditions on 2/5
Will case one, near full colonization with 50/50 peat/Verm
Last will be set into fruiting conditions at full colonization

i only case when growing penis envy and when i do i adjust the ph with hydrated lime. all other cube grows get a light top covering of coir at spawning.

I think I see bruising and a little bit of yellowish contamination... does that look like cobweb mold on top or is it just mycelium?

Just swiped 4 plates with a rusty whyte spore print I received from Caps. I'll post pics as soon as I see growth because honestly I'm going to need to be walked through this.. sorry guys. A noob has gotten his hands on this awesome strain.

Made a batch of MEA, PC for 45min @15psi
Poured in my still air box. I Used a homemade inoculating loop that was torched red hot between each plate. I left it overnight to cool and harden. First thing this morning I inoculated 14 Petri dishes
4 Rusty Whytes
3 Phoenix oyster
3 shiitake
3 king stropharia (wine cap)
1 that was not inoculated (control)

Any thoughts, advice, criticism is much appreciated.
