Cloning usually works fine for me with store bought mushrooms. I try to pick the youngest, least dry specimens for cloning. Best shiitake's for this I found were in asian grocery store, much better shape then regular grocery ones.
Never looked at portobello's closely, but I am sure you can find something that will work.

For Portobellos see if they have cremini, they are baby Portobellos and tend to be fresher and drop good dark prints

How should someone sanitize the donor mushroom before scraping flesh and transferring to agar? Just alcohol wipedown? Or soak?

Don't use alcohol. It soaks into the tissue and dehydrates/kills it.
I just brake the body apart vertically with my fingers, and biopsy a small piece from the newly exposed surface of the stipe.

Some people use desinfectants of some sort, I don't. Never had issues, even from mushrooms picked in a forest. Store bought ones are probably cleaner then that anyway.

not alcohol. that will kill the myc.  people usually use hydrogen peroxide, diluted bleach solution, or iodine. I've used all 3 and h2o2 is the cheapest easiest to obtain and use IMO.
