Signs of Bacteria in Grain Spawn

I've done a bunch of searching and still have trouble identifying when cube grain spawn is completely clean, slightly bacterial, or more severely effected.  Discoloration is easy, but what about when the mycelium just grows "weird."  There are a lot of responses to peoples pictures saying its bacterial without real explanations as to why that is the opinion.

Maybe a kind soul out there has pics of perfect grain spawn and slightly bacterial examples?  I use mostly oats and some rye. 

Or even better, a previous deep discussion on the topic (with pictures).

There's a thread full of contam examples  UTSF. For a practical approach if the grain isn't dry but growth becomes slow or stagnant with no visible mold it's probably bacterial.

Thats a great thread, but not exactly what I'm talking about.  Its easy to know if you have bacterial contamination when there are uncolonized areas of jar, excessive liquids, or different colors.  When you have nice white 100% colonized grains, but something is still wrong, those are the tricky ones.

Usually sweat on the glass... condensation looks a little different... can develop a thickness to it, look slimy: all the while completely colonized

Healthy mycelium is sorta transparent and lighter, u can sorta see the color of the grain through the mycelium.

Bacterial mycelium is thick and foamy looking, very dense and bright white.

But it's hard to kmow the difference till uve actually seen clean spawn. I don't have pics sorry, I'm sure someone does.

That's just it. It's more identifying the degrees of bacterial.

You learn to recognize the clean look after you've seen it put out several good flushes before it goes to shit.

Super bacterial will look kinda thick and will contaminate after the first flush or two.

If you shake your jar and it recovers slow (more than 24hrs) or grows back different looking, than it's so bactetial you should throw it out.

If ur trying to avoid bacteria, wash ur grains WELL b4 hydrating. Pc longer than normal. I do 2 hrs. 1.5 hrs I end up getting some bacteria, my pc is probly fucked up, and clean ur cultures on agar.

U'll never completely avoid bacteria, it's everywhere. But when it outnumbers other microbes, that's what's called a bacterial infection.

Think like vagina (sorry ladies). There's always bacteria, but when there's a high number if particular bacteria it causes infection. Basically the same thing here.

Well I use wbs and it's dirty as fuck. So I wash it real good to get what I can off it.

Haha Idk man, I do wbs with my eyes closed now. It would feel weird to switch. I stick with what I know.

Here are a few jars that I suspect are bacterial and/or contaminated.  The wispy growth inbetween the the whiter growth is a bit concerning.  Have a look and let me know.

But what's it being dirty have Todo with bacteria.

How would washing before hydrating prevent bacteria.i don't understand.


My wheats probably dirtier than your wbs.

They are yes... you're looking for that type of sweat on the jar, and the bright white, creamy looking mycelium; especially prevalent in the bottom of the right jar at the bottom... may also be mold on the left side center of that same jar... matter fact, pretty sure both of those jars have a grey mold of some sort, all 5 are bacterial, but I can't see any obvious mold in the first 3... not that it isn't there, just from the photo... are you using a SAB? Pressure cooker? Agar?

Well, its good to know I can at least identify there is a problem.  I did everything by the book.  Everything in a SAB.  Spores to agar, transfered  to new plates. Innoc'd with clean plates.  Hydrated and Pc'd grain jars with Bods oak tek.

I had a fairly high amount of contamination in my agar work, that I threw out.  And that can't be attributed to anything else but fucking up in the SAB.  So I am leaning towards there is where improvements are needed.  I wipe everything with iso, spray walls with soapy water, wipe everything with iso that goes in, flame every time I use a scalpel, no hands above plates or jars, etc….

There are some success stories (at least I hope). I grabbed a few random jars out of my good egg pile.  They look ok?






I also put some Tamps and ATL's on rye, and those all look nice and clean.  No contams or problems there….Maybe my oat prep is off?

Are you waiting 10 minutes after setup to begin? Elevating your plates? Using slow concise movements? I only wash my hands to the elbows with antibacterial soap and warm water, and I work my SAB dry... no gloves, no iso... I don't get many satellites, typically contamination only occurs in germ or tissue plates, and not always then...

Yep, I wait a while to start work and plates are off the floor elevated. I wear a long sleeve shirt with gloves.  I'm working on the movements, but am at least aware of what I'm supposed to do.  A few mistakes are made with cutting wedges and what not.  I definitely don't use anything I accidentally drop or touch a non sterile surface.

One possibility is that I inoculated with some whole plates or half plates.  Read somewhere about it being better to not use any of the agar that is towards the edge because of higher possibility of contams on the walls of petri.

Another possibility,  I use the SAB with wet paper towels as the floor.  The lid isn't flat and can't be used.  I know some people use wet towels, don't see much of a difference between the two.

Whats the word on that second set of spawn jars?  Big ups for the help.

I'd spawn them, still a bit of bacteria here and there, but nothing that concerns me... try drying your oats overnight

Are you venting your PC properly? Do you wipe your SAB with soap water? I would stop putting the wet paper towels on the floor, just be sure to clean the surface well beforehand. Are you reusing blades? I use a fresh sterile blade for each transfer and I sterilize the handle in the PC.

Just buy 2. Scapels and 100 blades you'll never run out. Plus u can flame the handle :awesome:

Don't vent for 10 minutes, vent for AT LEAST 10 minutes. You need a steady stream of steam, no air, coming out before you put the rocker on.

The point is expelling all the air and replacing it with steam. If you don't have a steady stream then you obviously haven't expelled all the air.

I can't help but observe that my rye jars have 0 signs of bacterial issues, and have problems in half of my oat jars.

I'm gonna prepare some more oat jars with the following changes:
1.  lids with SFDs instead of poly
2. rinse the oats before boiling
3. dry oats super well before loading jars
4. increase my vent time in PC

I'm about to do some G2G with what good jars I have and am nervous about having more contam issues in the transfer.  I have more rye too, gonna do G2G onto rye as well to see how it goes.  Im guess there is no issue with using an oats master to innoc rye grain.
