Thanks. Yeah I almost lost my APE and APEU. Working out finally...
Hey unc, dunno, i keep transferring and noccing grains and some shit works out. I ain't the level of precise recipe science that some of you guys are, so my recipe varies and even my go to LME/P is different, at least a bit, everytime. In fact, this week it''ll be 20g agar, 6g LME, 1g NY, 1g P to each L of distilled water. Of course those are targets and sometimes I spazz the addition of the powder, lose some control. [neuropathy ] I'll see how that goes.
Would you guys run the same recipe less the agar for LC broth?

I have some 4-year old swabs that don't work. Trying to get them to work. I PCed some distilled water, let it cool. Cut the tips off 2 swabs, dropped into 2 syringes. Sucked up .5 ml of the water into each one. Capped the syringes. Did all this in front of flow hood.
One syringe I am leaving as was, the other one I added a rubber band to give some compression.
Any ideas how many days I should leave them soaking before using? Thank you.

What's a good/reliable tek for modifying the traditional widemouth quart metal lids that'll only be colonized by agar transfers?

1st on looks ok to me, I think most people would do another transfer, definitely another transfer on the 2nd pic.
APEU swab plate and T1 both pinning

Perfect example of why you should take multiple pieces from a germ plate.

Meaning it won't work, or meaning it takes just hours or minutes if it's going to work?

It's gonna be fine. I think they're saying you don't need to wait. But, given that they're kinda old it probably doesn't hurt, probably is actually beneficial to wait several days. I mean, spore syringes are common and those are good after months of soaking.

Itโs pretty common to be pressure hydrate old spores like that. Definitely wonโt hurt.

Thanks all. I'll give 'em 4 days total.
Here's a happy little tree of Orissa mycelium. I just transferred away from 4 oclock and 6 oclock.

I just wanna say my last batch of 24 agar plates had 0 contams, a personal best. Perhaps it was washing my forearms and gloves. That was added to wiping everything down with a wet bleach towel, put under the rack.
Of course it all adds up, I move slower, especially taking the cover off a plate. I even pause after moving stuff around getting ready for the next plate.
I tried incense today and it's cool to see what air currents happen.
Incense is for temple!

Nice! I keep wanting to do the incense test but that stuff gives me and my wife splitting headaches for hours if we're around it. I've recently started wiping my forearms with iso and sterilizing my tools as well and have seen notable improvements with all my transfers
