So I’m still struggling to pour agar and not have a ton of condensation. I pour a sleeve per 500ml bottle and work from the bottom up and let them cook in front of the FFU for 24-72 hours before I re-sleeve them. I have a temp gun, and today I poured the first sleeve at 130, second at 120, third at 116. Had 2 broken dishes in one of the sleeves.
Also found my first contaminated plate today. A T1 tidal wave plate that was pretty ugly. The transfer was 9 days ago.
Here is my pouring setup and my ugly plate. It looks like the contaminated plate had the contam dropped on it by the condensation?

Are you putting them in the fridge? I've stopped doing that and get much less condensation.
I pour them, tape a single piece of tape around them in stacks of 10, put the stacks in the sleeves they came in, tape that up. I put them in the corner covered with a clean shirt. Let them sit at least a week to make sure no contam.
Then they can sit 4 to 8 weeks without getting too dry.
Don't put them in a warm grow room, just in a room temp room.
If you end up with too many and they start getting dry, pour less of them more often.

That's metabolites from contamination, if it was from the condensation it would be all over the plate.

Hey everyone!
I'm cleaning p.azurescens LC on agar. The plan is to create clean, fast growing culture. The reason why I don't want monoculture is a long growing/fruiting cycle, so I think some genetic variety would be beneficial.
First picture is 12 days ago. Outlined sectors are for transfer on new plates. Also, I'm thinking about using one of this sectors for inoculation.
Open for advice and opinions

I’m not putting them in the fridge. I leave them in front of the FFU to cool and let some of the moisture hopefully evaporate, and then tape them and leave them at room temp. At room temp they condense up as soon as I tape them even if they clear up while they cool.

That's metabolites from contamination, if it was from the condensation it would be all over the plate.
Thanks! This is an awesome lesson of the day. Glad I was at least right that it was contaminated as todays trash day and it’s already on the curb.

I'm curious about some of your guys typical agar procedures.. Do you guys save your old plates after taking transfers from them? Or do you toss them right away? What is your preferred method of disposing of old plates? When you go to grain, do you also do a transfer to another plate so you have a genetic copy to go back to?

When you say you are planning on using them for inoculation, do you mean taking them to grain? Those look like disorganized germ plates to me. You will want to take that a couple more transfers out before going to grain. Your highlighted transfer areas look good to me. Get them on some new plates and see how clean and organized they become.
I like to always wait till at least a T2 before going to grain. Usually a T3 though. Sometimes nasties can hide and become enmeshed in the mycelium on the germ plate and first transfer, and since this is a woodlover, you are going to be investing a lot of time growing this culture, so you might as well make sure it is anexic.
Don’t worry about getting a “monoculture.” You won’t get one through a few transfers. Probably not even after a few dozen. Even the tiniest transfer the size of a needle poke from your MS germ plate will contain a vast amount of strains and genetic diversity. Just focus on getting a clean culture to work with. That is the key.
How did you make you LC? Was it a clone, or MS?

I have a tendency to hoard plates, but after half my fridge was food and half dishes i decided to man up and start dumping way more.
Unless you're a pheno hunter i think it's perfectly fine to do 1-3 germ plates. Choose the best one, do 1 transfer to 1 plate then another transfer. Spawn. Choose clone. Make like 1-3 master plates and just transfer from those to new plates when you wanna make spawn. I dump all the ms plates.
If you're a paranoid bugger, it's good to think about that the more mycelium coated shit you have in your house the worse for you if you get snagged.

Took this pin off coir I spawned in front of the flow in a XLSA after steaming, took it off in front of the flow too.

How do you orientate the sleeve when you place it back on the stacks?
Presumably the sleeve goes over the sleeve.
Have you noticed if this reduces condensation compared to wrapping them individually directly after pouring/cooling?

I am a little health nut, i don't even own a microwave :P.
I made a "discovery" today. I am currently in the search of a perfect culture and then plan to stop fucking around with spores for a bit. I am also trying to find a way to streamline the process a bit.
I had a syringe. I went two routes.
a) normal syringe - agar - spawn - bulk
b) pf puck with a squirt of spore solution
I expected it, so not the biggest surprise, but maybe some newbies will appreciate it. The process of getting a clone from pf pucks was almost 3 weeks faster than the agar proccess. The benefit of pf puck pins vs agar pins is the fact that you can actually judge the quality of the fruits much better than on agar.
So just a quick tip: if you want a fast clone, try knocking up a pf puck.
