Where do you guys buy your sterilized petri plates? Im looking for 500 pack or hell, even batches of 20. I used one from Amazon and they arrived cracked and air was loose in bag.

I use glass. 20 x the cost but worth it. Am around the 12 x mark for now, and only broke 1 through stupidity.

I'm going through my second case from amazon, and the first case had maybe 10 cracked in 500, not seen any across 3 sleeves in this new case: different brand,  and packaged better... I didn't really sweat the 10 in the first case, as they're plastic, ~11 cents each, and likely delivered by a dope without a fuck to give ... I was kind of happy it was ONLY 10 lol

Fingers crossed for your outdoor bed. I laid down some pine bark last year and 'salt - baed' some spores (and spore water) on top. I hope mother nature helps out now.

Cool, I will try the water and see how that goes. I have tried placing a spore print in a 0.1% peptone water and placing on an agar plate with antibiotics... it grew a lot of yeast lol.

At the moment I am seeing blue mould on top of what looks to be another filamentous fungi, but I'm not sure if it's the blue mould before it changes colour (if that makes sense). I would like to cut a piece of the edge and transfer. But it may be safer to just try from the spore stage.

Thanks so much for that info, I will do it with my first finds of the season!

Those are what I bought, and some arrived cracked. One of the bags was almost at full air capacity, it was blown up like a balloon. Not sure if that means anything or not, but there was no consistency. Maybe thats the norm for petris everywhere though, I don't know.

Ease and speed,I had 3 fully grown out plates cut each into 10 and transferred to 30 plates with working in a SAB makes it faster. The 30 plates go to 90 grain jars then 45 mini monos and that's about max capacity for me.

APE isolation project I've  (evidently) started  (without realizing)... with appropriate sized wedges only!

14 are from the same parent plate: the 2 on top marked "R2" and "R4"(for rhizo 2&4)  (1&3 got tossed)(2nd and 4th plates, top row) taken on the 21... the 2 columns under them, and the last column (transfers from "R3" before disposal) are transfers from those 3, taken on the 28th (2 days ago)... let the culture war begin!

Smaller wedges might take a day longer... and will help ensure a clean culture with fewer transfers... also, don't have to let the plate fully colonize before use... I've been cutting entire cultures out for qts as small as ~1.5" in diameter for SAGARI drops  with no real discernable decrease in colonization speed,though I've been pouring them on the thicker side so there's enough agar to coat the grain thoroughly enough to maintain the effectiveness of the method... typically see full colonization in 5-6 days without the need to shake

I should probably have said that the fully grown out plates are from an established clean culture normally t3-4 from clones I take. And are "appropriate" sized :p

Gotcha... so why not just drop 10 wedges to qts and skip the last transfer? Just to have more future in the jar? Not really saving time, as these plates have to grow... just curious, I find all the differing methodology Interesting af... 45 2qt minis? Just for easier isolation of possible contaminants? Space? Like, as opposed to less, larger tubs?

Drop 10 wedges to quarts then g2g to the full 90? The math just worked out better in my head. I have the 90 qts and the 30  clean plates. it seemed more appealing to do that. I havent actually dropped the wedges yet. I also thought having "clean" agar for each individual jar would be better than g2g and spreading any possible contamination. Assuming there is any. It's my first go around doing anything of size. I have wire racks set up with t5s that fit the 45 minis perfectly and went with the minis to again minimize possible contamination instead of doing fewer larger tubs. Plus I think they're easier to handle in general, harvesting, maintaining(if any), moving.

Having more culture in each jar I thought would speed the colonization of each jar, though its probably not very noticeable comppared to having smaller wedges dropped.

I'm still pretty new to cultivating. Any advice, critique, and knowledge is always welcome.

I like LI when doing the final transfer, agar to grains. Instead of transferring individual wedges to each jar, I take one dime sized chunk and drop it in a quart jar with 2oz sterilized water. Then I give it a shake and suck it through the injection port.  Makes more than enough liquid inoculat so I usualy use extra, resulting in stupid fast colonization, or you can use it sparingly and make it spread a LONG way.

Interesting, that sounds a lot easier than what I was about to do. All my lids are fitted with injection ports already as well. With so many inoculation points with the Ll I'm sure it cuts down time to just 3-4 days to full colonization too. I'm definitely going to give that a go.

Sure does! Yeah just use a 1 piece lid with only an injection port, no gas exchange. You will notice a small vacuum will want to suck the content of the syringe back into the jar but it’s very easily managed.
