Have this plate of Caerulescens from 1/31 that looks like it’s trying to pin. Took a couple transfers but was thinking about swabbing some myc for an LC also. Think that would hurt my chance of getting it to pin?

It's just 9g agar, 2.5g +or- brown rice rice flour and 500ml water. That may be mixed with diluted grain water, not sure.

Don't be concerned with isolation, but you want a clean culture... this looks like it's carrying bacteria IMO ... id let it grow to see if the white will overlap the thickened clear leading edge and fluff up a little... I like running MS tubs as genetically variant as possible and pick clones for isolation... isolating on agar prior to fruit is a crapshoot in the dark... You can even get super lucky and isolate a culture that absolutely will NOT fruit...

Hi folks,
Just hoping to get a little advice on culturing what I think are P. subs.
Media: PDA + antibiotic Starting material: spore print on paper
Method: I had a pretty spore print stuck on my pin up board since last season, and tried to 'inoculate' my media using 2 methods.
1. Cut small sections from the print and place directly on the plate. 2. Scrape spores onto the plate in little 'piles'
All of this was done in my living room, but I figure the antibiotic should be forgiving... but I have fungal contamination, the powdery green centres look bad maybe Penicillium?. There is some clear/white small growth (top right hand corner). I would like to know if I can cut this out and place it ontop of another PDA plate.
Also, I was wondering if you can recommend any good peer reviewed resources for this type of work

I've had limited success with p.sub wild-spore-on-paper (newsprint) to agar.
Firstly I'd recommend reducing the number of spores. Off foil, a dab on print to dab in agar is enough. With paper, rub 2-3 times, preferably with your loop cooled in agar first to encourage it to be sticky. Less is more.
Secondly I'd suggest increasing your sample rate. More plates = higher chance of success. You can use multiple sites per plate as you have, try 5-10 plates.
Thirdly, keep it cool. These fungi thrive in the outdoors, and keeping them cool, even refrigerated can be the head start they need over mould.

Thanks so much for your reply Bobbit. I think I may try keeping a few prints on foil this season.
ok, got ya "less is more". I wonder if I can make a liquid spore suspension, and dilute a few times. Do you know if there is a liquid media that gives P subs an advantage over the green moulds? If not, I can just try the 'light dab' technique lol.
Sweet, I will try a few more plates next round, and place them in a cooler spot (at the moment, I have them in my laundry).
Have you tried using a section of the stipe on agar?

I did some stipe->agar->grains->cardboard->soaked woodchip->outdoor beds.
Being that this is my first season, I'm not sure they will fruit this season, though here's hoping.
As for spore solution, you could put one of your paper triangles (take a tiny piece) in a jar of sterilised water with 100ml or so. After a few days soaking, one drop per innoc. site per plate, in the cold.
Any nutrient water will work against you (I'm led to believe from lots of reading) agar+patience is your best bet.
Make more dishes that you intend to use in your initial set so as soon as you see myc, transfer to the empty plates . . .
Tissue from fruit gives you a very good chance of viable fruiting culture. In a SAB with a flame sterilised scalpel ready. . . Hold the base of the stipe in 2 hands and split without touching the freshly exposed sterile tissue. Cut a piece from the inside and flick onto waiting agar (only open the plate immediately before tissue transfer) close plate quickly. Wrap before removing from SAB.
You will see growth in 12-24 hours.
If you have a fresh syringe, and a fresh fruit, try the Josex Biopsy method. Use the stipe! One of my attempts was through the cap and ended up with multispore.

Quick agar question. When the myc hits the edge of the plate. Can I still use them to tiger drop ect. No pour pasty plates.

They look hella uniform even after taking 6 samples from it. I have a few older dishes from a few days ago and to think I been tossing all my old cultures to make fresh plates. Damnnnnnnn.
