It went from an MEA plate to an MEA plate with charcoal and yeast extract. The new nutes definitely tripped it up for a good week. It was so weird seeing the change from a ropey jump off to instant transparency.

Does charcoal act as a nute? I thought it was just for that goth charm. Someone said mycelium eats food coloring but not charcoal.

Do you think it was mostly the yeast extract?

Do you think it was transferring from one nute to another? If so, that's worth some experimentation. Let me know please what thread to sub if you start on that!

Do you think there is any chance this is APE myc? It's almost as high as it is wide, it's on a obviously very contaminated plate, it doesn't look like it should and it's not where I swabbed.... but it is uniform, it is eating the food dye and it is growing at myc speed, no quicker.

It looks nothing like healthy myc I've seen before but it's from one of the swabs Roarkell kindly sent me and I would love to save one so if there is even a 1% chance I'll transfer.

Thanks AKA it looks weirdly less suspect in the photos than it actually does. It's really fluffy and round. I'll definitely transfer away though and see what happens.

I have a general question about agar, do you have to use something with FAE to tape Petri dishes shut (micropore, parafilm), or can you use a non-porous clear packing tape?

Can knocked-up plates be stuck in a box in a closet, or do they have to "breathe" a bit?

Yeah a germ(ination) plate is the one you germinate spores on. In regards to the FAE, a lot of people use saran wrap/cling wrap on their dishes with great success. I don't see why packing tape wouldn't work, but it would be annoying to take off when you want to work with the plates. They technically do need to breathe a bit, but if they're growing slowly, the rate of oxygen consumption drops off pretty drastically. At low temperatures you can probably store them without any fresh air for a pretty long time, like a couple months maybe? I know a lot of people have good success storing cultures in the fridge long term.

Packing tape isn't flexible... I doubt it would work.

In respect to adjusting to new nutes... my agar drops are taking longer to leap off in my millet bags than in my wheat bags, could that be a reason? (agar & LME plates)

Millet grain soak water, you could probably put some millet in a food processor and use it instead of brf in brf agar.

I just saw that a local mill (bob's red mill) makes a millet flour. Could I try the same weight of millet flour in the stead of the LME?
