Hello all, new member here

I am going to be trying to do my first agar work to start this whole thing off in a week or so when everything comes in, I just need to get some spore prints to work off of ordered. I think I am planning on doing both pour and no pour to see what suits me. I am looking forward to starting this process.

Ive got some pp5 deli cups as well as I purchased some of the linked containers in this thread, to try out.

IS it just personal preference over PDA or LME or grain water agar recipes? I have materials to do all three. just curious for actual member to member confirmation. Thanks

I prefer proper petris all around. I use malt because it's cheap, readily available, consistent, and stores well. Haven't used PDA, and grain water just needs diluted properly.

I prefer LME over grain-soak water because I can get it to be more clear. I've never used potato anything but a lot of people here do. If you have everything you need I would start with LME or potato though since it will be several weeks or more before you get a culture to grain so you would end up just wasting the grain you soaked to make the agar.

Okay yeah originally I was planning on just doing PDA but went ahead and got some malt as well. And good point, I guess I will have to be isolating a few times, so yeah.

off topic from agar kinda but and to you all, so It doesn't really matter too much what spore print I start with correct? I know sellers like to label them but at the end of the day they are still spores withh varying genetics. Anything I should or shouldnt start with?

:whathesaid: you're already way ahead of a lot of people by starting with a print, instead of syringe AND learning agar to start. APE (albino penis envy) is a cruel mistress lol.

I've really enjoyed my Chitwan and Golden Teacher cultures so far. Very few mutations, good potency (though this is purely genetic) and a fun high.

Plate pins can be super quick dropped to grain or lc.


Just ask ol’ no plates faht.

T1 from the plate pins is giving me a lot of clear growth. I was hoping for thick white ropes now that I've switched to LME but no luck. I'm a bad photographer but with my eye I can see there's nice organized growth so I'm not super concerned. Maybe they'll thicken up after a transfer or two

Thanks been reading a lot, I felt like in the long run it would save a lot of time, by trying to start with prints and agar, and if I get contamination in a plate, I may be able to clean it up and transfer, or if I loose a few plates, its much less of a problem than loosing whole attempts at spawning and fuiting only to have to start completely over only to buy another syringe and risk it again. Idk just seems like that was a lot more work than learning a little right from the beginning??

And okay I will certainly stay away from that lol.

I've a slight issue where I can't send these to grain (if they're ready that is) so can I pop them in the fridge to slow them down, or can I take more transfers and pop them in the fridge?

There is no reason to lose a culture to contamination. There are always ways to get the organism you are after.

I work with a lot of wild fruits and spores. If it is alive you can clean it up.

Here is a sporulating mold ridden germ plate that cleaned up very nicely. It was clean in one xfer but it got 2. This is a very minor mold infestation IME and came from an indoor print...

There are a LOT of different maneuvers you can do to get the organism you are after. Lots of folks preach the fear of mold in one way or another and believe in old wives tales but I don't think they really know what they are looking at. Fear is dated tek.

It really helps to have a microscope or some way to look up close.

The only reason to ditch a culture is that it is not productive.
