I’ve had that happen with a couple cultures. No idea what it is. It looks healthy as far as I can tell but I was too weirded out to put some to grain.

I heat about 2.5” water in a pot to 140 and put my media bottles in there while I pour the other ones. It helps cool them faster and then maintain temp better.

Yeah see that separate disc of agar in the bottom is what seems to be sucking up all the agar. I bet if you reheat it the rest of it would be way thinner than it should.

My yeti, clones all did this after the first transfer. I hadn't thought anything of it and ran one anyway, without issue.

To what extent can you transfer away from bacteria/molds in agar?

I've recently been getting into it and started shitting myself every time I see contam plates, especially if a high rate. I know the point is to transfer away from contam, I just didn't realise it'd be such a high rate to start with.

At that point I'd been using spore syringes.

I've only just realised today, because I'm fucking stupid, that all that nasty mess must be from the syringes rather than my technique.

I mean, I already knew syringes weren't clean. But god damn every time I see the mean green and bacteria all over my plates I can't help but feel I've done something wrong.

Since I've started transferring away from cleaner plates, or working with spore syringes with no visible spores, I'm not getting contam so far.

I feel a bit better about it all now.

But yeah regarding my first sentence, as long as it's not meshed in with or growing over each other, I'm ok to transfer from nasty looking plates? Or would trich just spread their spores around?

You can work with pretty bad plates imo. So long as the piece your transferring isn't touching a tam then I'd go for it. With mold I wouldn't open the plate if its sporulating, or if your gonna try, do it last thing in the session, and clean everything after.

If the plates are bad it'll probably take multiple transfers before its properly clean.

With syringes you could try squirting a bit in a sterile petri and the using an inoculation loop to smear some on an agar plate. Reduces the amount of liquid on the plate substantially.

Thanks dude, I appreciate the fast reply. Got my clothes washing right now and prepping to get some more agar on the go.

Note taken on the swab! I've got one to try lol.

Some decent looking agar I can transfer from atm.

Mystery strain T3
PE T3
TOC germ plate
APER T1
PEU T1

Also... I've got a mystery strain T2 starting to produce pins. Would one be ok going straight to LC with some flamed tweezers?

I wouldn't. By all definitions you should be able to and be alright considering the plate and the environment is "sterile". But to my understanding a pin is going to have a larger surface area and won't visibly display any mold or bacteria the same way agar would. I would just put it to another plate to verify it's clean then go from there.

Never considered pin to lc, maybe if it was invitro but then your just making an lc of an untested clone.
I'm kinda on the fence, it should be fine but I don't see the point. But then again I dont really do lc cus I suck at it.

Lol, its not the lc its using it to inoculate with im not great at. But then again it was my very first attempt at pans and LC. So I suppose I was bound to trip up on one of them.
I do need to give it another go. I wanna switch to bags and lc would be far easier than g2g in a sab

Lc is worth learning to pull off. It’s just so fast.


I did a side by side with spores a while back, lc vs normal wedge dropping and I spawning the lc tub before I even got to grain the normal way.

Pour your lc. If you can g2g you can pour lc. I think its easier:shrug:

I usually swish mine around as it cools and if it solidifies I toss it in the microwave and heat it up to liquid again and let it cool for the second time. It’s annoying though..

look up 'hot pour' clean up technique. this is what you want to do

I'll do up another batch and give it a go.

With the pan lc i free poured about 25ml into my grains but there was a little pooling at the bottom and eventually they stalled. I tried this a couple of times to no avail.

I should try it with cubes tho, I know I can grow them at least, so that a better start.
Do you guys roll your jars a bit after nocc'ing with lc, or let them sit until growth occurs, then shake.
