Ya APE being a pain to germinate isn't uncommon. Pretty sure pasty said once it's clean slant it and never do it again lol

I’ve burned through 5 swabs and got nothing. A spec of bacteria that was it.

Thanks man! Wish I would get to using petris so I could take better pics lol. I think honestly I'm going to put that first Argentinian plate to some grains tomorrow since the growth is looking so solid and those KSSS i'm trying to semi-isolate the best growth until they are perfect.
I'm thinking all my KSSS plates i'll keep transferring till about 20 or so and then see what they are looking like. Probably not going to get a perfect isolate but we shall see. They are the UFO phenotype so I want to find the clone with the best saucers haha.

I've been getting really light and whispy tomentose growth on my plates. I used grain water as my nutrient base. I'm up to T6 on a few of my transfers and still little to no rhizomorphs growth. All of the isolations so far have been from MSS, as I'm still waiting got first fruits to clone something nice. I've now moved to LME and Agar (10g of each to 500 mL). Will this increase the likilihood of changing the growth pattern of the mycelium? Growth appears clean, but not as textbook as I see there. Also - I accidentally bought 47 mm plates. They obviously colonize faster. How long do you wait to make a transfer on average?

Depends on how the plate looks some xfer at dime size, I usually get around to it at silver dollar sized later xfers might not take place until the myc is getting close to the edge

Thanks Joze! I'll keep it updated as I keep testing some of these plates. I think I'll make 3-4 20qts with some of my best plates and see what goes.
And Rumple it seems to me that you aren't always going to get rhizo growth with every culture. I've especially noticed this with my KSSS, I have about 7-8 clones and not a single one has rhizo growth, all of them are pretty wispy but they still have been working well. I'm on T7 on about 5 of them and haven't seen any signs of that pretty ropey goodness. But tomentose growth isn't bad at all just sometimes makes spotting bacteria harder and not as picture pretty lol.

First attempt at using Agar plates. These are all germination plates.I had no clue what I was doing on the first few, then found C10's write up and did about 40 more plates using the streak technique outlined there. You can't see, but on some I used my full r-tard strength and tore the shit out of the agar (lol).
The TOC spores are from a TC (Thanks again), the UKC(Unknown Cubensis)is from my first monotub that I took prints from, and the Tx Syringe is from a local homie of mine. Definitely had some blue green weirdness on a few of the UKC plates and tossed them, hoping that some of these are okay to do some transfers in a bit, hoping for some input from ya'll

Thanks for the response. You as well, D3mon... If nothing else, I'm getting lots of practice in before I start with tissue cultures and Master/Slave management. Gone through 75 plates in 2 months just farting around with transfers and Tiger dropping clean agar into new jars. Lots of fun learning.

Tomentise myc usually comes from too nutritious of a recipe.
I would dilute your grain water or use less lme

Grain water works fantastically if you boil it and sterilize it proper. I dilute mine down a BUNCH with distilled water. LME agar is also a great choice. I do 2.5g LME to 500ml of distilled water. Everyone has their own recipe and some use low nutes for plates to get that really nice rhizo growth.
