Hey guys- still new here. Hoping to get some input from the vets on this plate.
When I made the initial transfer off the source plate prior to this, I was picking up a small cut of rhizomorphic growth. However, once transferred, the growth started looking more tomentose(or is it just a combination of weak genetics + less strain diversity present in the dish?) and created these ringed cross-sections. I'm currently at about 150ish+ hours accumulated of research, and I've read that these rings can indicate hidden contaminates, swings in temperature, or having been exposed to excessive light (Is that still accurate?).
My question is more of a general one: What do you guys see when you look at something like this? If I make another transfer, where would you suggest that come from? I see the V-shaped sectoring at 12o'clock, but what's with the weird divoting in the mycelium towards the edge to the right of that?
I'm at a weird mycological intersection of tons of reading knowledge but very little hands-on experience. Any input is appreciated!

Looks alright but you will want to get a couple transfers out like a week ago.

I haven't tried using just lme, so I'm gonna give that a go this weekend

You won't look back. . . LME has a fairly complex nutrient profile that keeps myc happy. . .

I stopped wasting my time with nutritional yeast. I'm lazy and don't want to strain so it just adds grit to my agar.
Haven't noticed any large difference in growth.

Thanks for the suggestion! I transcribed the agar recipes from the Radical Mycology book and put it in this thread: https://www.shroomery.org/forums/showflat.php/Number/26795122 I hope this makes the agar recipes more accessible and easier to find. And also encourages people to try new interesting agar variations.
There is for instance a beet root agar recipe in there. Would be interesting to see how it performs in comparison to the beet agar that RyeHumor made.

My bad no yeast for Malt extract. I need to one of these days be less lazy and play with my dextrose. But MEA works so great and I'll never go back to the days of grain water agar. (Contam rate was about 70% with grain agar, with just minimal MEA contam rate is basically nill)
The beet agar has been done on here within the past 3 months? I believe it was D3monic

I did a search and it was RyeHumour exactly 2 months ago. This post The recipe in the book is a MEA that has been supplemented with beet. Ryehumour used a water extraction without any additives. That's why I think it would be interesting to compare them.

Yea I pretty much exclusively use lme agar. Keep meaning to try grain water but it ends up in the drain before I remember

Second transfer of Amazon and B+ both looking pretty good! Planning to do some slants from these. Still quite new to the hobby, but excited for the success I've had with agar so far.

Well I still have tons of Oat left so I'm sticking with oats. So after Oat prep I pour the broth in a pitcher and deal with it after laying out the Oats to dry. So this is why I'm into Oat broth Primal's Experimental PE Grain Inoc>Agar Isolation Log (with pics and paragraphs) I find his premise interesting.

I've had great success in the past with Rye and Barley grain water. I dilute it by half and then add my agar for the equivalent ML of Water.
WBS agar always contamed on me.

I'm very new, but have been using strictly lme with much success. Grain water is appealing to try, although with having lme on hand I don't see any reason to try. I also have been running a slightly less nutritional mix, like 60/40 agar to lme as I read it makes the myc seek out nutrients more. Obviously, as I said, newbie approach and just trying things out but it made sense and so far is working well (except with swabs.. Thinking I need a higher nute ratio for that maybe?).
I would think when using grain water it would be a little more difficult to know proportion of nutrients to agar?
