You can use just a section, but the entire agar plate still needs to be clean of any contaminate.  The more sections you use give the grains more contact area and you will get quicker colonization (see Tiger Drop method). 

Some people believe you should not use the entire plate as there is a greater likelihood that there are contaminates on the outer edge where the condensate drops run off the container walls.

Try keeping a control plate that you don't knock up.  I doubt your contamination is due to your lack of a wrap.  What are you inoculating with?

Anyone ever try running many different recipes against each other at the same time?  I prepared the following recipes and did my transfers tonight:
  • LME 1.3%
  • LME 1.3% w/ food coloring
  • LME 1.0%
  • BRF 1.5%
  • Trypticase Soy

Anyone know where nutritional starvation begins to become noticeable?  I ran a bunch of dishes with 1% LME against 1.28% LME and culture growth was the same.

I know it can effect the growth and some people pour thick, but I just never felt the need. :shrug:




There was a side by side comparison post somebody did recently and all of the thicker plates grew considerably faster.  Try your own experiment to be sure.  Me personally, I've damaged enough transfers due to thin plates on the donor side and thin plates on the receiving side that I only pour thick.

I'm running a nutrient variable experiment right now.  Spent time over the weekend painstakingly making individual no-pours and precisely increased the amount of nutrients in each one.  The baseline was 10cc of water in each no-pour and 0.2g of agar.  Varied the LME from 0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0%, 3.5%, 4.0% and also BRF at 1.5% & 2.0%.  Tonight I carefully moved over the 10 transfers.  The dishes were slightly soupier than I would have preferred but I'm still really excited to see what they look like next week.

Will post a thread if it ends up panning out.

You know, right, that to produce any useful data from a "change one thing only" grow require using single culture clone selected down to a single sector and grown out.

Without that, there is too much variation possible with a multispore grow to really determine anything.




Yeah I don't actually plan on publishing this one to the Journal of Mycological Sciences, maybe next time though.  In my home lab there are obviously many other variables, sector selection, wedge size, accuracy of my measuring devices.  Regardless it will still give me a sense for what works better with the products that I use.

My nutrient variable experiment didn't turn out perfect, but I still took some results away from it.  It's very difficult making single no-pour jars and my least precise step was measuring the water using a graduated cylinder.  Each jar was supposed to have 10cc, but I would imagine some had +/- 1cc.  I think this caused some of the jars to have pools of water, which either carried the culture around or a contaminate. 

Things I found interesting:
  • My BRF is garbage and consistently shows extremely weak growth
  • My goto recipe of 2.0% seems to be the lower threshold of good growth
  • Higher percentages of 3.5-4.0% showed rhizo growth (could be a fluke based on sector)
