If you're pcing your agar in two separate bottles as I sometimes do depending on how much I need, then I'll microwave because how do you know you are getting the same amount of nutrients/agar in both bottles if half of it is sat at the bottom of the measuring jug?

Do the math per bottle.

Mines 3.75 grams agar 7 grams lme per 400ml and that's per bottle.

By the time you've weighed and measured everything twice mines is already finished in the microwave. So no real advantage to either way. Just personal preference I guess:shrug:

I want to make a big batch of LC now that I have clean agar plates to take wedges or biopsies from.

I tried grain water already but my order of dry malt extract just showed up and I wanna give it a go. It should be fine just with that and water right? I don't actually need dextrose?

What ratio? couple grams to a litre? or? And lastly, do you guys filter the LME broth so it doesn't have sediment in there or you just leave it ?

I'm Not doing agar. Just LC. I imagine the same amount of LME is probably fine though

I'll just experiment, lol and whip up 4 or 5 see which one does best. Gonna try 8 grams to 500mL see how it looks. If it ends up too dark I'll dilute. Sediment, half I'll filter half  I won't and see which ones do better.

On the agar subject, if you read good cooking guides for Agar they always say to use Cold water to dissolve it, as it's insoluble when cold, but disperses correctly. Adding to hot water result in lumps.

I only ever start with cold water and heat for a few mins as you get perfect consistency:thumbup:

Misread the post, my fault m8, I know nothing about LC, other than they are a great inoculation method, and are a great way to fuck up a pile of grain, mostly my experience.

If it takes some time for the dry patches to dissolve during the cycle you're cutting your cycle time short. At only 15m at 15psi if your agar dissolves during that time you should add time.

When I worked in lab at University there was media that was heat sensitive so we cooked it at 20psi for 3 minutes rather than 15psi for 15m. For whatever reason hotter for less time was safer for the media to not break down. But with only a 3m cycle you need to make sure youre homogeneous before running the cycle.

And with malt extract upon the first time it reaches boiling for a few minutes a bunch of proteins precipitate out making your media hazy as a final product. Unless you decant it off the crud first

I use a FP pre mix right now (for a year or so) and I just weigh it, mix with hot tap water in my flask and then PC for 45 minutes at 15psi. Works great every time.

My PC hardly stays at 15 it's usually in the 18-20 range so I think I'll be okay I've been doing it for like 500-600 petris.

Ate 3.5g last night of my first harvest of GT...Meh.  maybe grew some bunk shrooms :shrug:

I dont think it matters much. Some dishes have rings that lock them in place so stacks dont topple. My guess is these are the ones without

Is it normal for PE to take over 2 weeks to show knotting/pinning?  I don't have knots yet in my tubs and it has been 14 days.  The growth looks very healthy in all the tubs, though, and they have been fully colonized for almost a week.  I have done APE before successfully but not PE so I am used to seeing pins within 10-ish days.
