First Agar Work - Cottony Growth
    #25624792 - 11/19/18 08:30 AM (6 years, 1 month ago)

Hey guys, it's my first agar work. Inoculated 3 plates with B+ spore swab. 7 plates waiting in the fridge for healthy culture. Now my question is when should I isolate them? What is the yellow spot?

The satellite colonies are contams, the center is B+ mycelium. In a still air box, cut a piece of clean mycelium and place it on a new Petri dish.

Sector the fastes growth, make sure this portion is also uniform with no thin spots and what not.

Satellite
colony is the one that is on the edge of agar? So is is time to isolate or I should wait little bit more?

What do you mean by sector the fastest growth? Sorry, I'm really new to agar work.

Take a sample of the fastest clean growth free from the slower growth. Transfer this sample to a fresh plate of agar. Repeat again then either repeat and/or inoculate  grain.

Thanks for the reply. I did the transfers 3 days ago.  It seems this time growth might be rhizomorphic. Contams also began to show themselves on inoculated agars.
