Hello forum.  I am pretty new here and i have been mostly reading and absorbing the wealth of knowledge this forum has to offer.  I would first just like to say Thank you to the people who have made this possible.  This community is awesome and i am glad to have an opportunity to be a part of it.  What you all have created here is pretty amazing in all reality, so thank you for having me.

Secondly i am going to be starting my first go at this soon and i would like to run my choices by you guys and see if you have any pointers or suggestions to help me find the path to successful flushes. 

I am going to start with agar.  I could quite easily be wrong but i feel like there is a higher possibility of disappointment with PF and to many variables.  Is this a good choice?

I am going to use the little glad containers to start(i will be upgrading to glass petri at some point) for two reasons. 1.They are cheap,  2.I am not yet comfortable pouring in a SAB.  I was planning on doing my agar a week or so before adding spores to ensure i have it clean is this wise?

I wanted to do APE originally because they were an amazing experience, but i am a bit concerned about yield and growth rates.  Is there something you could suggest that has a similar experience but better growth?  Yield isn't necessarily as big a concern as quality but i really don't want to start with a slow grower.

I want to put a lot of my focus into sectoring and isolation.(once i get my sterile techniques down)  I find that super interesting so spores aren't a big concern at this point.

Going to start on grain.  Rye if it is available if not whole oats are for sure.  I am reading it is suggested 1 cup of rye per quart jar. Is that correct? is that pre-hydrated?  1 cup of rye just seems like it wouldn't be very much in a quart jar.

When doing agar->grain i see some suggest putting the whole plate in the grain some only use little pieces.  Clearly more mycelium in the seed will offer quicker colonization of the jar but is there a generally accepted size, ratio, or portion added?

Having a hard time finding a reason not to do a monotub. I am reading a 2-3:1 substrate to grain.  Seems like a fairly wide range is their any noticeable difference between 2:1 and 3:1?  Also is there a reason for 60:40 coir:verm?  Just sort of assuming here but i would have though verm would have more nutritional value, then again from what i know dealing with reptiles coir is resistant to contamination so had to ask. 


When i finally get a flush how do i properly harvest it?


When i get going is it cool if i get a Newb grow documentation going?  Don't want to take away from the people that know what they are doing but it might be interesting to watch my learning curve.

Agar is always a good choice.

APE is definitely going to be a steep learning curve but if you're start with agar and picking good looking growth for your transfers you should be able to pull it off. I would recomend growing another strain side by side.

The rye will expand to about three time's it's size so one cup of dry rye per quart should be fine.

If you use glad containers ala pastywhytes tek then there is no need to do them a week beforehand. They stay sterile as long as they stay closed.

Doesn't matter how much inoculate you use. If the culture is clean a whole plate is fine.

If you succeed cut at the base of the stem.

Yes document everything.

Right on thank you.

One other question came to mind after i typed this up.  If i order say APE spores from somewhere.  How diverse are they?  (I guess i am not really sure how to ask the question so pardon me if my wording is wrong) am i dealing with something that has been isolated down and cloned or will their be different strains of genetics to work with isolating and singling out traits?

Don't think you can get APE spores on a print.Also remove the vendor name from your post.

my bad done, i wasn't planning on a print i was  going to go with a syringe.  I've not seen a print either.  To be honenest im not really sure how to work with prints and i felt like a syringe would be easier just happened thetype i want to do do not come in print apparently

It's a domesticated strain for sure but every spore drop contains thousands of potential genetic combinations both with desirable genetics and undesirable ones.

This is why when you're using agar it's important to select growth that looks strong and healthy.

Once you get things plated you can post pictures and we'll help you ddcide where to transfer from. Remember, you only need a drop or two of spore solution for plates.

Sounds like i will pretty much stick to the plan then with a week shorter timeline.  Thanks again for the input.

I will definitely be giving oats a shot at some point.  I have worked with rye before a bit so i am more familiar with it. (pappy was a moonshiner) Just wasn't 100% on amount of swelling.
