Clones or prints?
    #25605713 - 11/10/18 04:46 PM (6 years, 2 months ago)

I am making the switch to agar and I want to pour plates and get a culture started on them in the next day or two. Am I better off using a spore print from a good performer from a BRF to bulk shoebox, or would I have a higher chance of success using a clone from a good performer on a currently growing shoebox spawned with wbs?

The third option, an unused MS syringe, is my least preferred option but may also be easiest. I was just thinking that having spores which came from mushrooms that grew quickly and were large, or cloned mycelium, would put me a step ahead on getting a monoculture tub running.

Congrats on getting into agar. I would try all 3 if you wanted to. You only need 1-2 drops from a spore syringe to be sufficient. How long did you print your caps for?

I have two caps that I printed using Bod’s Tek (aluminum foil tent) for about 12 hours (overnight). I have two fruits in the wbs shoebox that are kicking ass and running out of names to take.

I have 10 mL of B+ MS.

10 mL is enough to inoculate 100-200 plates lol. Syringes usually last awhile.

Try all 3 for experience, but cloning is the way to go. Don't worry about monocultures.

The only thing i would try to work on is selecting a potent clone. If you get multiple clones going you can ensure you don't get stuck with a wimpy set of genes.

Cloning is the quickest and less contamination prone way to go IMO. Pick your best fruit, split it lengthwise down the middle, and take a sample from the stipe (stem), close to where the stipe meets the cap -at least that's the way I do it.

You can easily contaminate a clone tissue sample. Hence the advice to do all the options, and gain all the experience.

I am going to attempt all 3.  Maybe maximize my chances at success. It seems fairly straight forward, nothing I didn’t learn how to do in high school science.

That’s usually the hallmark of impending doom, though. The projects that seem easy are the ones you spend your whole life working on.

Thanks for the input, everyone. I appreciate it.
