[Help] Agar Prepration Question

hi everybody, i'm very excited, after all i want to give agar a try, i brought 'agar bacteriological' (which seems to be agar agar), and Malt Extract from a lab equipment store.
on the malt extract it's written: a water soluble extract of malted barley suitable for use in the cultivation of yeast and moulds content and consequently is very sensitive to over heating.

on the agar bottle it's written 30g/l and on the malt extract it's written 15g/l.

i was researching online and saw some people use these two and some other mix yeast with it. what should i do? do i need to buy brewers yeast or these two is enough? any suggestion for a good tek?

I only mix agar and malt extract, never heard of adding yeast. I mix them at 2% for each so for every 500ml water I add 10g agar and 10 malt.

I would make sure you agar is straight agar and dosent already have some type of sugar added to it already.

Your agar should be fine, considering you got it from a lab supply. Ratio as stated above is a good place to start. I like to add 11g of agar and 8-9g of LME and 500ml H2O, but as with everything in this hobby, YMMV.

awesome! thank you for responding fast, can i use tap water or it needs to be distilled water? as far as i found out, i have to mix them and pour them in a bottle and ps cook them for 40minutes @ 15psi, isn't that overheating for malt extract? (what they have written on the bottle and i have mentioned?) then i'm gonna leave it to cool down a little (1.5h) after that i will move them into my sta and pour them into my petri dishes. any correction needed?  :takingnotes:

I always use tap water never had a problem. Your process is correct as long as you plan on heating the water first. If you don't heat the water before you add malt and agar you'll have a hard time dissolving them.

i did it, that wasn't a total success but i managed to do it, my SAB floor isn't leveled so few of plates have agar on their door. how ever few others are just fine i'm gonna work with them and try to come up with something for the next time,

however i have a question, almost all of them has mist on their door, i moved them to refrigerator but still have mist. what did i do wrong?

Did you wait for the agar to cool enough? Did you stack them as you poured them? Condensation isn't uncommon but stacking the plates as you poured helps with condensation. I know it's basic but if you follow RogerRabbits strategy you'll have good plates everytime. Also takes a little bit of doing to learn.

yeah i watched that as you mentioned and i suspected i didn't wait 2 hours for agar to cool down, i was worried that it comes solid in the bottle, i will try again and update you on that.

another thing, i didn't buy parafilm to cover my petri dishes i used micro tape instead is that fine or have you any suggestion on that?

i tried to clone one of my mushrooms which i just harvested, cut in into two peaces and removed small part from center top and put it on my petri dish. i noticed that the remaining part of my mushroom turned green, is that normal, i cut it with a scissor. here's a picture:



and also why is my mushroom caps cracked? :/

Ya agar has to cool (generally in the PC for awhile) that's why all the teks do it.

Well that end of the clip I showed you shows him rewrapping them in the sleeve so there's that. Other than that I always use parafilm. Not to be a dick but are you sure you wouldn't be more comfortable using no pour agar teks? They don't use parafilm and you don't have to pour them.

Mushrooms crack because it's way to dry in there. I can tell that from the pic too looks dry as hell. I'd call that blue but blue has kind of a greenish tint to it sometimes, cubes bruise blue-ish.

point taken, no i like agar now. it's a bit strange because it's my first time doing it, i will get buy parafilm soon as possible, thank you for the details you provided.

sooo the green/blue parts are normal no worries?

Ya blues not always straight blue, looks weird sometimes. I'd worry more about how dry that tub is.
