Liquid Culture need hole in lid?

If im going to be making a honey and water LC, do I need to have a hole in the lid for FAE or can it be sealed?

I don't have much experience with LC but I thought you needed to have a hole in the lid for later, as in putting the needle through so you can collect LC in your syringe.

I am not innoculating using a spore syringe, going to be cloning with pieces cut from the inside of a stem.

You should first get into agar if you want to make LC.
Also you can only tell if a LC is contaminated or not by testing it on agar. ( or wasting prepared grain )

All good, ive done this TEK before years ago and it worked, just trying to refresh my memory on the hole in the lid.

yep you need some FAE but i wouldn't do it with polyfill or micropore.
i prefer 0.22ยตm filters

It's not a reliable method. You got lucky. Use agar.

You can do it with any filter, but make sure you don't get them wet when shaking. Polyfill will pass through contaminants if it is wet by your LC.

No you don't. FAE has nothing to do with this. It'd be for GE, but the primary reason is so you don't pull a vacuum.
