Really hard to get good pics due to the container and amount of condensation.

The agar in the highlighted area is no longer translucent and is cloudy and appears to be raised above the surface of the clean agar.

Bacteria? Or condensation?

There is a lot of condensation in these no-pour PP5 containers and I've incubated them at a slight angle so the surface moisture pools at one edge, rather than being dispersed across the centre of the plate. That edge coincides with the discolouration in this case. None of the other 9 plates show this discolouration and the agar remains clear and clean.

Looks like precipitate from where the condensation has dried up, just take a transfer from the edge of the mycelium to a new plate, aim for 3 generations before moving to grain

Thanks, does the initial inoculation of agar from spores count as one transfer?

This plate represents spores -> agar -> agar, so another transfer to agar is advisable?

Both transfers to date have been on PDA, should I change the agar recipe for the third transfer or stay with PDA?

It's up to you, but I germinate on agar then usually go 3 generations from that, I just personally feel more comfortable that there's nothing likely to be hiding by then

Yes if you've found a recipe that works stick to it..you can rotate around MEA, PDA or Oats etc, but it'll work fine
