my rye won’t colonize

okay. i did the tek
let it hydrate for 24 hours
then i boiled it for ten minutes. i didn’t have gypsum so i added powdered egg shells..
it’s been weeks and no sign of life
and this has never happened to me before
my guess is the grains were treated with fungicides or whatever... i just got my organic untreated rye 1lb for test and my calcium sulfate powdered!! so i’ll try again !! i will never stop  muuuahahahah!!
my money is it was bad grains
does that matter ?
if they’ve been treated or not

Yes it does matter...some grains are treated with anti fungal agents.

and it was also advertised as mushroom substrate and it’s clearly not.

it’s just been sitting there looking perfectly fertile and nothing

but i got some organic rye today, i’ll just dump out all 5 pounds of pointless rye :mad2: i mean

every time i use grain it’s been organic  but it has germinated atleast by 5 days

read rule #3's spores usually do germinate
and i even made my own spore syringe with a print i made from the last batch. i’m just starting with grains. i have bird seed



i just picked the rye and gypsum up today
going to try once more

anyone suggest a good wbs tek ?

Contaminated spores do not always germinate. Spores that are not shaken up well do not always germinate (because they don't make to the substrate). Old spores do not always germinate. If it's got the word 'usually' in it, it's not a rule :laugh:

sorry to hear you are having bad luck..

What tek is this tho? 10 minutes to boil grains? that seems like it achieves neither sterilization nor pasteurization effectively?

Not all contams are very visible, yeast can smell but is otherwise not as conspicuous as other contams for example.

Before you try to draw conclusions I would try it again but with a method that actually uses proper sterilization or pasteurization and continues with the substrate accordingly.

With grains you should treat it as a nutrient-rich substrate which means you must sterilize and maintain sterile conditions throughout inoculation and colonization.

How long to PC may depend on the size of your containers since the heat must penetrate into the core.

so:

- first wash the grains until no foamy stuff and residue is washed out..
- then do the 24h soak
- drain well and mix in supplements like gypsum
- put in a container which is well sealed and fit with a proper filter like jars or bags
- PC for 80-100 minutes depending
- let it cool completely (like overnight)
- inoculate etc

By the way iin general it is not a good idea to just inoculate grains with a spore syringe in my opinion because it is unlikely to be clean enough - should first be cleaned up on agar. But I guess cubes are so aggressive that people get away with this all the time.

To help being able to shake up a spore syringe well you can include a few drops of glycerol in the demi water you sterilize to make a syringe. Other things like lube can apparently work well too..

I've seen rye teks that say soak 24hrs and get to a full boil for 10 mins. Though it's been pointed out that you don't need to hydrate your grains so they "don't burst" or to "hydrate endospores so they're killed easier in the pc cycle". Also, I'm pretty sure the PC time is 90 mins, though to be safe I aim between 90-120 mins. My guess is it is the fungicides that messed you up because something should have grown from a spore syringe. No print is sterile so no syringe is sterile. Also, try using oats, you can get 50lbs for under $15. Just get triple cleaned and make sure they have the hulls still
