Semi-professional shiitake cultivation

Hello everyone and greetings from Spain, I communicate through a translator, I will try to translate as best as possible, I just established a shiitake cultivation company, soon show images of the facilities,I will show all my advances and doubts. Right now I am making bags of spawn, but the mycelium becomes very thin, the bags are 100% colonized but they are hardly appreciated, why? Thanks in advance

Then you might have either a mold or à bacterial problem. It should be much whiter after two weeks.

The wheat was cooked for about 15 minutes, until it was cooked properly, then it was allowed to dry until it did not get its hands wet when it was touched and put it in vacuum-sealed bags.

You can have bacteria without visual evidence. What is the smell? 1 TBSP per bag is not very much... smell should be similar to fresh shiitake mushrooms

Thank you, I did not smell, I know the smell of mycelium and grain with bacteria, this afternoon I'll put my nose to work. Should I put 2 tablespoons? Or what percentage?

I can not smell anything after the filter, I put pictures today. Would you use this spawn to inoculate the final substrate, if when you open the bags you smell of pure mycelium?

The other bags have this filtration system with 2 microns. Could I filter too much for shiitake mycelium?

Did you buy a mother strain from mycelia or a bag of grain spawn? My best guess is contamination in G2G transfer. Either in flow hood or due to insufficient sterilization of grain.

We will see it soon, but I think it is due to some other factor, such as that the humidity of the grain is not correct or that this strain colonizes in this way

You can not incubate wheat at temperatures higher than 22 C in spawn bags because of endospores. For jars you can go a little higher than that, max 26 C.
Also if you shake often shiitake can look like this. I believe you meant 3790. Might be okay, it does not like plain wheat so much. Try adding some sawdust to the next batch.

When we talk about cold or warm weather strains, we refer to fruiting temperature, not incubation temperature. If I were you I would incubate the bags at room temperature (arround 21ºC). You also have to be careful reading "incubation temperatures" because it's not the same "room temp" than "substrate temp". The temperature inside of the bag colonizing usually is 3 to 4ºC more than the room temp, so when you read somewhere that incubation optimum is 25ºC, you should manage the room temp to be 21ºC or so, in order to mantain the 25ºC inside of the bag. You can reuse the bags. The filters are usually good for reuse but the plastic is harder and you must be more careful sealing them. It must be clean for a good seal or it breaks easily. It might be easier to use new ones, buy them bulk and they will be cheap. I try to reuse when I can, because it is ecological.

You can make the incline steeper and have more agar in your tube. They will store for much longer that way.

A tube only holds so much. The more you tip it over the less it holds. If you lay it completely flat it is empty. Laying you tubes higher will make them hold more agar.

Better use sawdust by the way, you'll rip open your spawn bags with these chips and the yield is not going to be much better than sawdust.

Why couldn't you just PC some karo water, draw into a sterile needle, squirt it into test tube and draw it back up? Blasting it off and spreading the culture. I imagine you could even use a injection port cap on testtube.Easy to use, very reusable.
