Agar second transfer ?

This is my second transfer from multispore syringe how big should I let it grow before inoculating, jar? If I read right there’s no need for third transfer if second is clean, I think it appears clean but don’t know how big to let it get before cutting a chunk out?  I uploaded 2 pictures the second dish is it a contam growing on left side?  or is this just a different type of myc growth..  thnaks

There is a name for the growth at 7 o’clock to 11 o’clock on that pic on the right, it’s suspicious. You want all spikes! Left looks good, no reason. TO NOT make more plates for later right?!?

Well I have others similar to the left,, the pic on the right I just put there for feedback, since it looked funny , my other plates can grow out and save, I guess my question is how big of a wedge is normally used for a quart grain jar?

The second pic, the growth on the left side is called tomentose. It is several strains all on top of each other. The ropey looking stuff is called rhyzomorphic growth. Many people seek that out, but in reality it does not matter so much whether its rhyzo or tomentose.

I usually just let the whole plate colonize and then throw it in the jar since I typically make many plates. I have cut them in quarters before too.

Ok thanks I’ll let it grow it out the rest of the way and do 1/4 per plate they are 100mm plates I’m switching to the no pour mini plate tek, after this  I hate these big things

I like to make up a 60mm dish for each jar I plan to inoculate, and drop a whole plate to each jar. I've been mixing the agar soft  (7-8g per 500ml of water) and cutting the plates in a tic tac toe pattern, effectively making 9 (and sometimes 16 with 3 vertical and 3 horizontal cuts)wedges ... shaking the hell out of them after SAB session, basically liquefies the agar, and acts  as an LI essentially... full colonization in 5-7 days depending on variety

I love me a big ol petri. I get like 6 nice wedges out of each and then g2g each of those if they colonize clean. Super efficient.

You could always go a bit further and make more transfers to further isolaste.  This won't guarantee a better grow..  But that isnususlly the idea of plates to isolate and find good performers which u can store and come. Back to.

Yoiu don't need much for inoculating jars.  A small wedge is nolenty

man I’ve been using 20g agar to 400ml oat boil water wasting so much of my agar ,,  guess
I need to cut back on that to will last me twice as long. Had no idea so much less works lol

Damn thats a lot. haha Try between 8-10g per 500ml.

I Do pasty plates, I can make like 20 of them with 1cup(236.588ml) of water and 5g of agar.

Yea so  glad I posted this I killed two birds  it’s wih one stone,  was using this bodhista agar recipe 
” Grain Soak Water agar(save water from hydration of grain spawn add 20g/L Agar-Agar powder””
but for whatever reason I was thinking 20g per 400ml..    Just now realize it’s 20g per liter.  Thanks all for help/advice.

How do you like the grain water agar?  I have been using PDA, but thinking about making grain water agar and throwing a couple hardwood pellets in some for my wood lovers

The only other mix I have used is LME and the only different I can tell is the tint of it, growth is same for me
