Agar difficulties - BRF versus grain water agar
    #25383272 - 08/12/18 10:09 AM (5 years, 7 months ago)

Hi all, need some advice.

I have some dishes with BRF agar that had worked great.  Made some dishes of grain water agar that turned out a bit more...goopy than the BRF did.  I’ll also note I used Pyrex plates for the grain water and had used plastic plates for the BRF. Mycelia growth on the grain water agar is slow and almost see-through.  Pic is of the BRF (bottom) and grain water (top), with isolates taken just a few days apart.

Any thoughts on why I’m seeing better growth from the BRF plates?  I’m considering just using that for the time being since it seems to work better.

Grain water strength depends on lots of factors. Like dilution or how much grain to water you used in the first place. Boil/simmer/soak duration. Etc..

So any suggestions on how to dial it in?

Or, for someone who is still pretty new, should I just stick with BRF for the time being?

Really I'll say stick with BRF agar for now especially if you have gotten used to it.
On the list of agar , grain water agar is first and BRF is second(don't ask me way) so its not that big a deal. Also grain water agar is good at hiding contams sometimes.
Hope this helps you out:goodluck:

how can grain water agar be crystal clear and completely see through while simultaneously hiding contams exactly?

Food coloring is good at hiding shit but people like to use it to help see I guess. Imo nothing is better than transparency for helping see problems on agar
