Real Confession time.

This past month I have been trying to get "back to basics" and go back to following teks note for note. The reason being is that for the past 6 months, after the bad bout of trich/green mold that wiped out 60-80% of my growing efforts I was only using teks that involved as little open air time as possible My methodology was: Petris - 3 transfers of only the cleanest myc -> mycelium water -> my glass w/metal SHIP (no GE) lids -> LI/LC ->Grains ->Shoeboxes because I'm too scared of one bad jar of spawn ruining a mono.

These are the Jars in question:



The problem was, the last 4-5 batches(read OVER ONE HUNDRED) of my Ezi LC jars were not germinating/transferring properly, possibly a 90% fail rate. Yes, I'm ashamed it took that much effort to realise something was not right. The jars that did work would be 50/50% chance of either being healthy, fluffy myc (which would transfer fine) or spots/one blob of myc that would just stall out.  This wasnt the case before, before I had around a 60% success rate with them, not great but if you do a lot (30 a batch) thats still a LOT of workable, clean myc.

I blamed this on the age of my last batch of LI (some dated DECEMBER 20 FUCKING 17) but this was quite frankly embarrassing. What I should have been looking at is the gross vacuum that forms in these jars that make innoculating them a pain. I got over that by essentially just poking through the SHIP for a split second, letting in a jet of inoculate that the vacuum would pull for me. The problem started soon after I got my new PRESTO 23q, my previous PC was a shitty 10litre PC that only hits around 11-12PSI. The newer jars probably have a much stronger tendency to create and hold a strong vacuum.

So a few days ago I did a test of these jars, larger 250ml ones that I used for LC. However I wanted to see if this vacuum was the problem. I picked up a thread from Mycotopia:shake: a while back on AIRPORTS : empty syringes filled with pollyfill, plunger removed, thin fresh needle, and mouth taped with polyfill. PC'd with a batch of agar, unrapped, flamed and inserted in the ship to "Vent" the jar with heavily filtered air (takes a couple seconds.)

The results?

1/11 of the unvented jars have transfered. Like I said, my success rate was like 10% so thats consistent.

The vented "Airported" jars? 5/8 so far. 60% success rate. I'll definitely be adding GE ports to the jar lids. Poly/MP tape.

I confess, I feel like a fucking moron.

Fuck lc why bother? Just transfer from plates its a bit slower but much safer and less work. And an error source eliminated.

I broke up with my girlfriend half a week ago and am preparing my third grow ever right now. Partly to get over it, partly because she never liked that hobby of mine.

Got 4 pftek jars steaming in a pot, gonna innoc later this evening or tomorrow morning :syringe:

I've been out of the loop for two years, so I'm following shea25's 'pf tek done right' instructions, any advice or recommendations?

That tek IS very comprehensive, I must say. Up to (and including) point 4 I guess I managed to get everything right, now it's waiting time again :tongue:

I already went shopping for supplies once, because PastyPlates somehow caught my attention. Didn't have the time to get them going yet though.

I had considered tripping hard on 4g, lemontek'd and all, to take my mind of things. The past few days lead me to the conclusion that the breakup is for the better of both of us, so the schedule will be to just listen to music and stuff on 2g or less, since I'm long due anyway. The shrooms've been calling me for the longest time xD

I confess, I just scrapped my attempt at agar! I need to invest in a bigger PC as the one I have is not big enough for the amount I am trying to make and it boiled over! Might give some pasty plates a go tomorrow but am thinking I will have to wait till my next grow to clone and later next week to streak some prints to agar.  :ohwell:

I confess...... I forgot to shake my syringe before inoculating my master jars. I've lost sleep over it for weeks. They all ended up growing out just fine, but still. I kinda scared myself, it's such a basic mistake and I totally spaced out on it. My minds eye keeps playing back that huge glob of spores just flopping out into the jar. This event is fueling my desires to get into agar and isolates instead of depending of the luck of the draw in a commercial spore syringe.
