Transferring from plate with sporulating trich?
    #25332486 - 07/17/18 11:12 PM (2 years, 9 months ago)

Hello all!
I have an agar transfer question. If i open this plate, is it possible to transfer over the myc without catching some spores? This plate is the only one out of 6 that is contamd and has the strongest growth (of course). Can i open this in a glovebox and transfer to a new plate with any success or should i abandon hope?

Also thinking about adding a crapton of lime to the agar mix to keep any spores from germinating.

Thanks!

Don't add lime, that's stupid.

You could very carefully transfer away in a SAB, yeah. But if you have other clean plates, use them even if they're not quite as good. Maybe transfer that one as well, but get something clean to use so you don't have to start over. You'll likely have to do several transfers to get it clean.

Also, this isn't advanced.

These are the TEKs I use. They're all as cheap and easy as possible, just like your mom.

Anything you try to do to cover those spores will only fling more of them onto the desired mycelium that's there.

Personally I would toss it. You have other plates for a reason.

Adding lime sounds ridiculous. Also inhibit your mycelium from growing or growing well.

I have multiple plates, but mostly different species or strains of king stroph I try to keep in situations like this with success transferring away from sporulating penicillin.

Create a small cardboard sab away from your normal work area and do a transfer in there if youre paranoid. Two hand holes it a box.

Open the container slowly to not get air rushing in to move spores.
Transfer immediately and move your hands slowly.

If you get some spores maybe you can get some growth off the side  or before the mold sporulates and you can attempt to cut it out of the next plate(s) you transfer to.
