Slow Colonization
    #21607565 - 04/28/15 05:09 AM (9 years, 22 days ago)

Having a problem with slow colonization using the WBS tek. After P/Cing the grain, I inoculated the grain with my spores and put in a box with a heat mat to leave to colonize. The problem is the mycellium growth is taking a very long time. It will stop colonizing after 1/3 and then start back up again after a little while very slowly. Quartz jars that I inoculated in February are just fully colonized now.

The temp in the box is 27 C. I'm unsure what I'm doing wrong. Also some of the tubs that I inoculated have taken so long to grow any myc they are also getting other types of bacteria growing on them.

Any ideas what I'm doing wrong?

You made 2 commenly applied mistakes:
1) you use a heating pad, you should heat the room if it is realy cold. Otherwise just colonise at room temperature.
2) Spores do not germinate optimal on seeds, try going from spores>agar>grain>grain to grain>bulk

You could also have a bacterial infection. Can you post a picture of the jar?

80f/27c is a little on the hot end.

Did you shake the jars?

Toss if any contams pop up.

My wbs grow at done at room temp. Started with the original 3 pints and now I'm up to 10 pints of colonized substrate that's after spawning bulk and G2G.

MS > wbs

Wbs > bulk (spawning bulk)

Wbs > rye (G2G)

here is the jar I inoculated in feb   here is a tub I inoculated in March

As you can see there is only a little myc in the tub from march. I was using the V Tek if you are familiar with it! I have had success with only 3 tub so far as the colonization is taking so long. Do you think I should get rid of the heater completely?

Yes ditch the heater. 1st pic looks bacterial, probably why you're seeing it in your tubs is from using he heating pad. Bacteria thrives at higher temps. Next time shake your jars at about 20-30% colonization and they will finish faster. As above, it's best to inoculate with agar wedges rather than spores.

Best luck
