yeah, i mean, you're probably not going to get all too intricate in a 35 mm dish.
the idea of streaking is two fold
A: to ensure that you get growth in multiple places that are seperate from one another. this betters your chances of being able to easily transfer away from any contams.
B: dilute the spore count as the streak goes on. this limits genetics transferred and greatly speeds up the "thinning of the herd" in the isolation process. the more elaborate streaking patterns are designed to make this happen but any old streak will do it somewhat.

Again, you can literally apply the spores in any way you choose

How long does it usually take for agar to colonize after you squirt it with a syringe?

What's the protocol on reusing these? Ie.- Throw away if there's mold/bacteria in the plates. - reuse then if they were clean?

anything that's going back in the pc doesn't need thrown away after contamination. just wash and reuse. even things that don't get cooked don't need tossed, like tubs.

When I'd get mold on a plate/s I'd set those apart and next time I'd use the PC I'd simply put them on the lid and that heat alone liquifies the agar and it's enough to kill the bastard. Most people just clean them moldy plates normally and that's it, I'm just reluctant to let the bitch live after fucking with me and spread its spores to boot.
Also not a fun of tossing moldy jars, that's so wasteful, when all it takes to kill that shit is putting them in the PC and killing the heat as soon as it hits 15psi. Even this is overkill, mold dies way sooner than that.

Bleach isn't needed or wanted for anything really. Use soap and water

I made 12 pastysplates the other night, I had two stacks of six the ones on the bottom all have a noticeable amount of condensation while the six that were on the second row have almost zero condensation. I started the PC on low and worked my way up to pressure by slowly moving the heat to medium-high I let them rest over night in the PC to cool. Is there anything I am missing or is this correct I can add pics later when I get off work. PS I had them on top of the rack that was raised with two rows of quart jar lids.

I do the same raising them with two rows of jar rings and then placing them on the rack. A little bit of condensation is normal I’ve noticed. Shouldn’t effect how the plates work to much. At least it didn’t for mine. Sounds like you’re doing it right be allowing the pressure to build slowly. How do the napkins on top of the plates feel when you pull them out? If they are not to moist it could just be condensation inside from the heat. Usually goes away after sitting for a while.

The paper towels were wet not soaking but definitely wet, they have dried since then I've been working 13 hour days so no agar work yet but the have been sitting on the kitchen counter for three days and show no signs of contamination. I feel very good about having followed the ten correctly, couldn't find glad brand so I got the ziplock 2 oz squares. Used 1/4 inch holes covered in micropore. Then papertowels and foil.

Making up my first batch of plates for edibles/medicinals, had planned to do it last Friday but man...today is my last day off for fucking weeks.
I have Shiitake (spores), as well as wedges of Blue Oyster, Red Reishi, White Button Agaricus, and Cordyceps that I plan to put to agar today.
I'm very excited as I am new to edibles and medicinals on agar. Probably going to spend more cultivating these than I would cultivating actives.

Anyone have any ideas what made the agar splash all over the inner sides of my plates during PC'ing? I don't see that in anyone else's pictures? It must've been boiling like lava

I don't see the same thing in other's pics. Makes it really hard to look inside.
I followed the tek with kitchen towel on top and foil but PC'ed well over one hour because my PC only does 12psi. Also left them inside overnight to cool down. I did bring up the heat a bit fast I guess... Is that the problem?

The plates were sitting on the trivet elevated out of around 2" of water btw.
If I were to bring up the heat more slowly, does that include venting on low heat? In Bod's Pressure Cooker Use TEK it says to start by venting on high heat and thats what I followed.

I would think even if you brought the heat up to fast it would settle before solidifying. I don’t understand why it stuck and solidified to the corners like that. I think bringing the heat up to fast causes condensation but idk about what you have.
