Agar Cooling time before MS inoc?

Couldn't find an exact answer to this by search. How long does Agar in a dish usually take to cool before one can inoculate with MS? I plan to inoculate mine as soon as possible... they're just sitting in the SAB now. B+ and Penis Envy coming UP!

as soon as they're cool enough. if you can tell me exactly how hot the agar was going into the dish, your ambient temperature, how high the stacks were, how much volume you poured in to each dish. well shit I still couldn't tell you a time. just check them

How do I check without opening them? Tilt them? Should I be concerned the surface temperature will kill the spores?

You should be able to tell just by holding them how warm they are. I’ve only done agar plates a few times but usually for me about an hour or so after they’re cool.

What temperature are we looking for? Room temp? Dish is still warm?

When I pour thin and stack 10 high they are solidified in about 20min, but say 30 to be sure. Also depends a little i guess on the stiffness on the agar. I've been pouring very loose agar lately about 0.7%
And I pour at 45-46C

Depends mostly on the temp it's being poured at, thickness of the Puck and if they are stacked. Sometimes if you pour 4-5 stacks at once it will keep the Temps up for a bit longer inside the SAB.

yep an hour after pouring I start working on transfers, I also pour around 47°C on the temp gun. I just judge they should not feel warm to my touch when I begin work and then all will be well basically
not very scientific I know, but serving me alright so far
