A Scale-up Grow of Panaeolus cyanescens





I’ve been starting most of my grows with agar work, to make sure spores are clean and to select strains for downstream work.
I have some pretty clean prints now so I decided to skip the cleaning/selection on agar and directly inoculate from spores.

Acknowledgement: Blue Helix- This grow was partly adapted from  Blue Helix's inspiring Panaeolus grows. 

Path: Scraping from Spore print->Liquid culture-> Sterilized substrate->  Trays-> Recovery-> Case & Fruiting.

Materials / Recipes:
Print: Panaeolus cyanescens Australian variety

Liquid culture: 120ml: 0.4% Light Malt Extract, 0.4%(w/v) corn syrup brought up in grain soak tea in a 1/2 pint mason jar with a stir bar.

Liquid culture to substrate: 60cc syringe, 14g needle for inoculation of substrate with liquid culture:


Substrate preparation:
Horse manure 712g
Straw 38g
Millet 250g
vermiculite(coarse) 106g
Gypsum         1tbsp.
H2O 1900g ( to reach field capacity)
Total volume~4.4L split evenly in two large spawn bags. Pressure cooked 4hrs @15psi. The substrate was left to cool to RT and aerate for 12hr. then it was kneaded well to break up all clumps before inoculating. This volume is enough for two, 4 quart glass baking dishes.

Case: 50% peat moss:50% vermiculite, CaCO3 to pH~7.5.

Fruiting conditions: Greenhouse- 79-82F, saturating humidity, air movement-light breeze.

Procedure:
Inoculation of liquid culture:
Scraped a spore print with backside of a sterile scalpel to lift up some of the spores. Dabbed the loose spores with a sterile cotton tipped applicator and stirred the spores into LC.
Stirred on stir plate at low to moderate setting, temperature ~75-80F. for 12 days:


Liquid culture to substrate:
Inoculated ~50 ml LC into two spawn bags. The inoculated substrate was kneaded well to uniformly distribute the inoculant. Bags were incubated 13 days ~75-80F:


Substrate to trays:
Broke up the colonized substrate and laid it into trays. Mycelia was allowed to recover for three days in greenhouse- ~79-82F & saturating humidity:


Casing the trays:
Trays were cased to a depth of about ~1/4 inch. and incubated as above.

Fruiting:
First pins appeared 5 days after trays were cased. Shrooms grew to harvestable size and started dropping spores 4->5 days after pinning.
After harvesting the first flush trays were soaked in water for 30 minutes-> back to greenhouse. 2nd flush pinning appeared less than 24hr. later.






Traits of this Pan cy Aussie strain: Mycelia was relatively wispy, there was no problem with overlay.
This batch produced delicate willowy shrooms, in fact stipes were so thin that some of the shrooms were on the verge of tipping over.
Good spore drop produced nice dense prints. The 2nd flush produced ~1.5X more shrooms than the first flush and, as is often the case, the 2nd flush growth was more synchronous.
Potency?- I dont know- I haven't tried them and may not. If they are similar to the other Pans i've tried id expect at least 3X stronger than the average cube trip.

I didn't bother with any more flushes simply because the greenhouse (running for months) was turning into an algae factory.

Not speaking specifically of MS, IMO genetics are especially important to Pans.

Its my “semi” dialed in GH. Not saying this is the best way to do a GH but its served me well for several Pan grows- i'm not good at giving a short answer:
With the inherent limitations I have in the house:
I have the GH ( just one of those mini greenhouses with a clear zippered plastic cover) in a closet;
Air comes in from below and mostly goes into a wicking humidifier that has a pretty strong fan that blows air at ~80-85% humidity from below. The air escapes from an opening at the top.

I like to feel a light breeze with my hand in the area where the trays are in the GH.

Sorry - its just a feel thing- no real precision there. Also I want to state at the beginning- none of what i’m saying here is necessarily relevant to growing cubes altho I doubt it can hurt.

The air flow from bottom to top is from the fan in the wicking humidifier and a couple of rheostated appliance fans. Here’s the ugly picture of the bottom of the GH:



Thing is, there is an inverse relationship between air passage thru the GH and maintaining ~100% relative humidity- at least in my setup.
I neglected to say that I also have an ultrasonic humidifier that I use to up the humidity to ~100%- fog liked conditions at ~80F temperature. The ultrasonic is slaved to a humidity controller set almost as high as it can go.

So for me, I have to weigh the fresh air flow (movement) around the trays- (which I believe is important for optimal primordia formation) against the very high humidity conditions that also encourages primordia formation. 

Of course you don’t need this level of precision to get a "good" Pan grow- there are plenty of monotub grows that attest to that; but  imo a properly dialed in GH will outperform a monotub grow.

Hi My Colorado- as usual- I appreciate your thoughtful response.

I really dont like rich subs-
ie. the presence of BRF or other flours, worm castings , or other special ingredients EXCEPT for small scale PF tek, MacMerdin Pancake style grows where density issues of the substrate are less of a problem.

I played around with trying to scale up PF tek substrates- problem is the sub becomes too dense, heavy and  mycelia colonization is slowed way down- slowed down to the point that contaminants can win the race for colonization.

For larger scale grows- a simple sub with a high herbivore poo ratio. I think of key importance is the lightness- friability is very important. A light sub allows fast colonization by the mycelia- not by contams. I also prefer horse poo to cow- not because of nutritional factors but simply because horse poo is generally less dense than cow poo.

I didn't always think this way- but you live and learn.

This is Blue Helix's exact method. Same LC recipe as well. And this is the first grow that
I've seen that comes close to his results.
